摘要
目的在大鼠减体积肝移植模型中联合应用特利加压素和FK-409,检测术后移植肝组织中IL-10和TNF-α浓度及相关调控基因表达,了解联合应用特利加压素和FK-409对移植肝保护作用的相关机制。方法使用30%体积供肝行SD大鼠同系肝移植,根据不同的干预手段将受体大鼠分为特利加压素处理组、FK-409处理组、特利加压素处理联合FK-409处理组和对照组,术后第10天取移植肝组织,使用酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)试剂盒检测IL-10和TNF-α,使用逆转录定量PCR (reverse transcription-quantitative Polymerase Chain Reaction,RT-qPCR)和Westernblot检测炎性因子相关基因Bax,Egr-1,ET-1,NF-κB表达水平,统计各处理组炎性因子及相关基因水平。结果在减体积肝移植模型中,特利加压素联合FK-409处理组与对照组比较,移植肝组织中IL-10和TNF-α浓度降低(P <0.05),炎性因子相关基因Bax,Egr-1,ET-1,NF-κB在转录水平和蛋白表达水平较对照组降低(P <0.05)。结论特利加压素联合FK-409可有效降低炎性因子相关通路基因表达并减少移植肝组织中IL-10和TNF-α含量。
Object To study the protective effects of trivasopressin and fk-409 on the transplanted liver by detecting the concentration of il-10 and tnf-alpha and the expressions of related regulatory genes in the postoperative transplanted liver tissue by combining trivasopressin and fk-409 in the rat model.Method We performed liver transplantation using 30%volume liver graft in rats.IL-10,TNF-αconcentration and inflammatory factors related gene expressing level in liver graft were detected using ELISA and RT-qPCR after operation day 10.We divided recipient rats into 4 groups including terlipressin treatment group,FK-409 treatment group,terlipressin and FK-409 treatment group.ALT,total bilirubin,portal vein pressure and survival rate were detected in each group after operation.We compared results of 4 groups and found potential mechanism which protected liver graft injury after reduced-size liver transplantation.Result In reduced-size liver transplantation model,terlipressin and FK-409 treatment group showed lower IL-10 and TNF-αconcentration(P<0.05 and lower expressing level of Bax,Egr-1,ET-1,NF-κB P<0.05 and longer survival time P<0.05 compared with control group.Using terlipressin and FK-409 could reduced liver graft injury after reduced-size liver transplantation.Conclusion Terlipressin and FK-409 could reduced IL-10,TNF-αconcentration and inflammatory factors related gene expressing level in the liver graft,which contibuted to liver graft protection.
