摘要
目的检测肺癌组织以及细胞系中lncRNA DICER1-AS1的表达情况,并结合临床数据分析其临床应用价值。方法收集39例经病理诊断确诊肺癌患者的组织样本;培养肺癌细胞系A549、H1299以及二倍体人胚肺细胞系KMB17;实时荧光定量PCR检测lncRNA DICER1-AS1mRNA表达量。结合临床数据比较不同患者的lncRNA DICER1-AS1表达差异。结果肺癌组织中DICER1-AS1表达量0.98 (0.53,1.72)高于癌旁组织0.64(0.40,0.92)(P <0.05)。肺癌细胞系A549以及H1299的DICER1-AS1表达量分别为(23±2.7)、(42.3±0.48)均高于KMB17的(5.84±1.07)(P <0.05)。而不同性别、年龄、肿瘤大小、TNM分期与DICER1-AS1在组织中的表达高低间差异无统计学意义(P>0.05)。结论肺癌组织中DICER1-AS1的表达高于癌旁组织,其高表达可能会是促进肿瘤发生以及发展的重要因素,有助于临床肺癌对患者的诊断以及治疗。
Objective To detect the expression of lncRNA DICER1-AS1 in lung cancer tissues and cell lines,and to analyze its clinical application value with clinical data.Methods Tissue samples from 39 patients with lung cancer diagnosed by pathological diagnosis were collected.The lung cancer cell lines A549,H1299 and diploid human embryo lung cell line KMB17 were cultured.The expression of lncRNA DICER1-AS1 mRNA was detected by real-time fluorescent quantitative PCR.The differences in lncRNA DICER1-AS1 expressions between different patients were compared with clinical data.Results The expression of DICER1-AS1 in lung cancer tissues was 1.6±2.3 and higher than that in adjacent tissues(0.7±0.39)(P<0.05).The expression levels of DICER1-AS1 in lung cancer cell lines A549 and H1299 were 23±2.7 and 42.3±0.48,respectively,which were higher than 5.84±1.07 of KMB17(P<0.05).There was no significant difference in the expression of different gender,age,tumor size,TNM stage and DICER1-AS1 in tissues.Conclusion The expression of DICER1-AS1 in lung cancer tissues is higher than that in adjacent tissues.Its high expression may be an important factor in promoting development,and it is helpful for the diagnosis and treatment of patients with clinical lung cancer.
