基质细胞衍生因子(SDF)-1对人类伴糖尿病牙周病患者的人牙周膜干细胞增殖、分化影响及其可能机制的研究

摘要 目的针对伴糖尿病牙周病患者群体,研究基质细胞衍生因子(SDF)-1对人牙周膜干细胞增殖、分化影响及其再生修复的可能机制。方法拟从伴糖尿病牙周病患者的炎症牙周组织分离培养鉴定牙周膜干细胞,100 ng/mL SDF-1的培养液添加与否分为两组进行比较。通过MTT法实验方法来检测细胞增殖能力;Real time PCR检测碱性磷酸酶alkaline phosphatase(ALP)表达水平来确定细胞的分化水平。结果 MTT法显示P.g+diabetes+SDF-1组中人牙周膜干细胞增殖能力提高;PCR结果显示P.g+diabetes+SDF-1组中,炎性牙周膜干细胞中的ALP mRNA表达明显增高(P<0.05)。结论对于伴糖尿病牙周病患者群体,基质细胞衍生因子SDF-1作为重要的细胞因子诱导并促进了人类牙周膜干细胞的增殖和分化,起到类似中间衔接和催化的作用。
