摘要 目的研究6-姜酚治疗实验性自身免疫性脑脊髓炎的可能性及有效性。方法应用髓鞘少突胶质细胞糖蛋白(myelinoligodendrogliaglycoprotein,MOG)免疫C57BL/6小鼠,诱发慢性复发-缓解型EAE模型,观察6-姜酚对EAE小鼠模型的淋巴细胞浸润及TNF-α、IFN-γ、IL-1和IL-6等细胞因子分泌的作用,通过神经功能评分及病理分析等探索6-姜酚治疗EAE的有效性。结果 6-姜酚可抑制MOG诱导的免疫应答反应,通过改变TNF-α、IL-6等细胞因子的分泌,显著改善EAE小鼠神经功能评分,使发病时间延迟(P<0.05)。结论6-姜酚可有效缓解EAE,为6-姜酚用于临床多发性硬化的治疗提供实验依据. Objective To explore the possibility and effectiveness of 6-Gingerol in the treatment of experimental autoimmune encephalomyelitis(EAE).Methods The chronic relapsing remitting EAE induced by MOG35-55 in C57BL/6 mice were treated by 6-gingerol and observed for the changes of clinical signs and cytokine levels in the serum.Hematoxylin-eosin(HE)staining was used to observe the inflammatory infiltration in the spinal cord tissue.Luxol fast blue(LFB)staining was utilized to detect demyelination changes.Results 6-gingerol inhibited the lymphocyte proliferation responses induced by MOG,and by promoting the secretion of IL-6(P<0.05),it significantly improved the clinical scores of EAE mice and delayed the onset time of EAE(P<0.05).Conclusion 6-gingerol can inhibit the development of EAE in C57BL/6 mice,and can play a potential role for the treatment of EAE.
