MSTN敲除对肌肉增强因子2(MEF2C)和miR-222表达调控的影响

摘要 为脂肪沉积转录调控通路的研究奠定基础,通过生物信息学、Western印迹和RT-qPCR等方法研究肌肉抑制素(MSTN)敲除后对潜在调控脂肪沉积的肌肉增强因子2(MEF2C)和miR-222的表达调控作用。结果表明,在miR-222启动子区域预测到MEF2C的结合位点,Western印迹显示肌肉抑制素敲除后MEF2C表达量上调,qPCR结果表明当MEF2C高表达后miR-222表达量上调3.48倍(P<0.01)。说明MSTN敲除后引起MEF2C高表达,进而促使miR-222表达量上升。 The regulation effect of MSTN knockout on expression of myocyte enhancer factor gene 2(MEF2C)and miR-222 was studied by using bioinformatics,Western blot and RT-qPCR methods to lay the foundation for research of fat deposition transcriptional regulation pathways.Result:The binding site of MEF2C can be predicted in miR-222 promoter region.The Western blot shows that MEF2C expression is up-regulation after MSTN knockout.The qPCR results show that miR-222 expression increases by 3.48 times(P<0.01)after high expression of MEF2C,which indicates that the high expression of MEF2C caused by MSTN knockout can promote rise of miR-222 expression.
