摘要
对牛蒡子配方颗粒制备工艺筛选及牛蒡子苷元质量分数进行测定,采用正交优化方法进行处方筛选,HPLC法测定牛蒡子苷元质量分数.色谱条件:色谱柱:Dionex C_(18)(250mm×4.6mm,5μm);流动相∶甲醇与水的比例为40∶60;流速:1.0mL/min;检测波长:280nm;柱温:室温.结果表明,牛蒡子颗粒的最佳制备工艺:物料比即牛蒡子提取物、糊精、淀粉比例为3∶6∶1,润湿剂:85%乙醇,干燥温度:65℃.在上述色谱条件下牛蒡子苷元在1.41μg~7.05μg范围内线性关系良好,r=0.999 9.方法回收率100.09%,RSD为2.19%.该方法可为牛蒡子相关制剂的开发和质量研究提供参考.
To screen for the optimal preparation process of Fructus arctii formulated granules and to determine the arctigenin content in them.The orthogonal optimization method was used to screen the prescriptions and HPLC was performed to determine the arctigenin content.Analysis was carried out with Dionex C 18(250 mm×4.6 mm,5μm),the mobile phase was CH 3OH-H 2O(40∶60),the flow rate was 1.0 mL/min,the wave length was 280 nm and the temperature was room temperature.The optimal preparation process of F.arctii granules was as follows:The ratio of F.arctii extract∶dextrin∶starch was 3∶6∶1,the wetting agent was 85%ethanol and the temperature was 65℃.Under these conditions,it had a good linear relationship,the average recovery of arctigenin was 100.09%,and RSD was 2.19%.The method described above can be used for the development and quality control of F.arctii preparations.
