摘要 为开展柑橘种质资源抗病性鉴定和评价提供技术支持,采用Folin-Ciocalte法测定柑橘叶片多酚含量,以柑橘叶片多酚提取含量为指标,利用正交试验法分析乙醇浓度、液料比、提取温度、提取时间对提取工艺的影响。结果表明,柑橘叶片多酚的最佳提取工艺参数为液料比50∶1,乙醇浓度30%,提取温度40℃,提取时间30 min。该工艺成本低、易操作,可用于柑橘叶片多酚的测定。 To provide technical support for the identification and evaluation of disease resistance of citrus germplasm resources,the polyphenol content was measured by Folin-Ciocalte method with the extraction rate of polyphenols from citrus leaves.The effects of ethanol volume fraction,liquid-to-liquid ratio,extraction temperature and extraction time on the yield of polyphenols were evaluated by orthogonal test.The results showed that the optimal extraction process parameters of citrus leaves were 50:1 liquid-to-liquid ratio,30%ethanol concentration,40°C extraction temperature and 30 min extraction time.The process is low in cost and easy to operate.This process can be used for the determination of polyphenols in citrus leaves.
