瓦布贝母与松贝母中贝母素甲和贝母素乙的含量对比研究 被引量:1

摘要 目的建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)同时测定瓦布贝母中贝母素甲和贝母素乙含量的方法,并与松贝母进行比较,为科学评价和有效控制瓦布贝母药材质量提供可靠方法。方法色谱柱:Dia-monsil Cl8(250. 0 mm×4. 6 mm,5μm),流动相:乙腈A-0. 05%——二乙胺水C(70∶30),流速:1 m L/min,ELSD漂移管温度:85℃,氮气流速:1. 5 L/min,增益值:2,柱温:30℃。结果贝母素甲和贝母素乙分别在0. 452~2. 712μg和0. 660~3. 96μg范围内线性关系良好,加样回收率(n=6)分别为99. 50%、100. 37%,RSD分别为1. 14%、1. 91%。瓦布贝母栽培品中贝母素甲和贝母素乙的含量均高于其野生品和松贝母。结论此方法操作简便,结果准确、可靠,可用于瓦布贝母药材的质量评定,瓦布贝母栽培品中生物碱含量较高。 Objective To establish a method for the determination of the contents of peimine and peininine in cultivated Fritillaria.wabuensis.,to study comparative analysis comparing with Fritillaria sungbei,to provide a reliable method for evaluating scientifically and controlling effectively the quality of Fritillaria wabuensis.Methods The HPLC conditions:a Diamonsil C 18(250.0 mm×4.6 mm,5μm)column was used,the temperature was 30℃.Acetonitrile-water containing 0.03%diethylamine(70:30)as the mobile phase,with flow rate of 1.0 mL·min-1,the ELSD was used as detector with temperature for the detector drift tube of 85℃and the nitrogen flow rate of 1.5 L/min,gain of 2.Results The linear ranges of peimine and peiminine were at 0.452~2.712μg(r=0.9998)and 0.660~3.960μg(r=0.9985),the average recoveries(n=6)were 99.50%and 100.37%,with RSD 1.14%and 1.91%respectively.The contents of peimine and peiminine in cultivated Fritillaria wabuensis were higher than the wild product and Fritillaria sungbei.Conclusion This method was simple,accurate,reliable and can be used for quality assessment of Fritillaria wabuensis.It provides the advantage basis of high alkaloid content for being received in“Chinese Pharmacopoeia”(The first of 2010 Edition).The contents of Alkaloids in cultivated Fritillaria wabuensis are higher.
