摘要
目的通过基因芯片技术探测HBV感染对肝癌细胞耐药相关基因表达的影响,为深入研究HBV在肝癌多药耐药中的作用及机制提供新的依据。方法以HepG2细胞作为对照,用稳定表达HBV的肝癌细胞HepG2. 2. 15模拟HBV感染的肝癌细胞,利用基因芯片筛选出耐药相关差异表达基因。Real-time PCR检测部分耐药基因表达以验证芯片筛选结果的准确性。结果肝癌细胞HepG2和HepG2. 2. 15中差异表达基因共1 263个,与药物相关基因58个,其中上调基因数31个,下调基因数27个,涉及药物转运相关基因、药物代谢相关基因、细胞增殖相关基因、细胞周期调控基因等。Real-time PCR检测MDR1、MRP2、LRP和GST结果与芯片结果一致,确定芯片检测结果可靠。结论 HBV感染影响肝癌细胞耐药相关基因表达,为进一步研究HBV感染在肝癌多药耐药中的作用提供线索。
Objective To screen the effect of HBV on drug resistance related differentially expressed genes in hepatocellular carcinoma cells by DNA microarray. Methods HepG2.2.15 and HepG2 cell line were resurrected and amplified in vitro. The differentially expressed genes were determined by microarray. The significant differential genes were selected by KEGG (Kyoto encyclopedia of genes and genomes) database. The drug resistance genes levels of MDR1, MRP2, LRP and GST were further verified by Real-time PCR. Results In HepG2 and HepG2.2.15, a total of 58 drug resistance related genes were considered differentially expressed. There were 31 up-regulated genes and 27 down-regulated genes. The differentially expressed genes were involved in transportion, metabolic, proliferation and apoptotic signal pathways. MDR1, MRP2 and LRP were up-regulated with significance while GST had no obvious alteration at mRNA level by Real-time PCR. Conclusion HBV affects the expressions of many drug resistance genes in hepatocellular carcinoma cells.
