摘要
目的观察微小RNA-299-3p(miRNA-299-3p)对人肺癌细胞A549迁移、增殖、凋亡、多柔比星敏感性的影响。方法以A549细胞为研究对象,首先评价miRNA-299-3p对靶基因三磷酸腺苷结合盒E1(ABCE1)的调控效应;通过CCK-8染色、划痕实验、活力分析、TUNEL染色以及蛋白印迹方法系统分析miRNA-299-3p转染的A549细胞迁移、增殖、凋亡以及对多柔比星敏感性的影响。结果 miRNA-299-3p对肺癌细胞ABCE1具有显著抑制效应。经miRNA-299-3p转染的A549细胞增殖活性、迁移等生物学特征并未受到明显影响。然而,miRNA-299-3p显著增加了A549细胞对多柔比星的敏感性,与对照组相比,同样剂量的多柔比星药物导致A549细胞活性显著下降,凋亡水平显著升高。结论 miR-299-3p靶向抑制ABCE1表达,其过表达并不能显著抑制A549细胞的增殖,但miR-299-3p联合多柔比星促进了A549细胞对多柔比星的敏感性,显著促进了A549细胞的凋亡。
Objective To investigate the effects of microRNA-299-3p(miRNA-299-3p)on the proliferation,migration,apoptosis,and doxorubicin sensitivity of human lung cancer cell line A549.Methods The human lung cancer cell line A549 was selected.We firstly investigated the regulating effect of microRNA-299-3p on the target gene of ATP-binding cassette E1(ABCE1);CCK-8 staining,Scratch test,dynamic analysis,TUNEL staining,and Western blotting were employed to analyze the influence of microRNA-299-3p on proliferation,migration,apoptosis,and doxorubicin sensitivity of microRNA-299-3p transfected A549 cells.Results MicroRNA-299-3p had significant inhibitory effect on ABCE1 gene of lung cancer cells.The basic biological characteristics of lung cancer cells treated by microRNA-299-3p,such as the viability,proliferation and migration,were not significantly affected.However,microRNA-299-3p significantly increased the sensitivity of A549 cells to doxorubicin.Compared with the control group,the same dose of doxorubicin significantly reduced lung cancer cell viability and increased apoptosis level in microRNA-299-3p treated cells.Conclusion MicroRNA-299-3p inhibits ABCE1 gene expression,and its overexpression dose not significantly inhibit the proliferation of A549 cells,but miR-299-3p combined with doxorubicin promotes the sensitivity of lung cancer A549 cells to doxorubicin and significantly promotes its apoptosis.
