红景天苷对大鼠脑出血后神经功能的影响 被引量:3

摘要 目的通过研究红景天苷对大鼠脑出血后自噬相关蛋白的影响,探讨红景天苷对大鼠脑出血后神经功能的影响。方法将120只清洁级SD(Sprague Dawley)大鼠随机分为正常对照组、模型对照组、假手术组、红景天苷组、红景天苷+3-MA组、红景天苷+雷帕组,采用Bederson评分法对各组大鼠进行神经功能评分;采用蛋白免疫印迹法检测大鼠脑出血后自噬相关蛋白LC3I、Beclin 1、Cathepsin D、P62的表达。结果各组大鼠治疗后的Bederson评分相比,红景天苷干预各组均低于模型对照组,差异具有统计学意义(P <0.05),其中红景天苷+雷帕组评分最低,红景天苷组次之,红景天苷+3-MA组评分最高;脑出血大鼠模型的自噬相关蛋白LC3I、Beclin 1、Cathepsin D、P62对比,红景天苷干预各组均表达均低于模型对照组、假手术组及正常对照组,差异具有统计学意义(P <0.05),其中红景天苷+雷帕组表达最低,红景天苷组次之,红景天苷+3-MA组表达最高。结论红景天苷在大鼠脑出血后具有改善神经功能的作用,其机制可能与调节细胞自噬有关。 Objective The effect of salidroside on autophagy related proteins after intracerebral hemorrhage in rats was studied to explore the neuroprotection effect of salidroside after intracerebral hemorrhage in rats.Methods 120 Sprague Dawley rats were randomized into 6 groups:control group,model group,sham operated group,salidroside group,3-MA plus salidroside group and rapamycin plus salidroside group.Neurological function scores of rats were examined by Bederson score.Expressions of LC3I,Beclin1,Cathepsin D,P62 were detected by western blotting.Results The Bederson score of three salidroside intervention groups after treatment were significantly decreased than model group(P<0.05).Among the three salidroside intervention groups,the Bederson score of rapamycin plus salidroside group were the lowest while the 3-MA plus salidroside group’s were the highest.The Expressions of three salidroside intervention groups’autophagy related proteins were significantly decreased than model group,sham operated group and control group(P<0.05).The autophagy related proteins’expression of rapamycin plus salidroside group were the lowest while the 3-MA plus salidroside group’s were the highest among the three salidroside intervention groups.Conclusion Salidroside might improve the neurological function after intracerebral hemorrhage in rats,and the mechanism might be achieved through the inhibition of autophagy pathway after intracerebral hemorrhage.
