小檗碱调控线粒体功能改善2型糖尿病引起的肝脏脂质代谢紊乱 被引量:11

Berberine Improves Lipid Metabolism Disorders of the Liver Caused by Type 2 Diabetes via Regulating Mitochondrial Functions

摘要 目的探讨小檗碱对糖尿病肝脏病变的作用,及线粒体参与其中的相关分子机制。方法实验分为3组:正常组(WT),糖尿病组(db/db)和小檗碱治疗组(db/db+BBR)。HE染色观察肝脏组织形态改变;检测血浆总胆固醇(TC)、总三酰甘油(TG)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)的水平;Western blot法检测肝组织线粒体相关蛋白的改变。结果与正常组相比,db/db组中TG、TC、LDL-C的含量明显增加,而HDL-C含量明显降低,ALT、AST蛋白水平也显著提高;与db/db组相比,小檗碱治疗组各指标明显改善,差异具有统计学意义(均P<0.05)。同时,小檗碱显著改善糖尿病性肝脏引起的空泡化。此外,小檗碱激活线粒体自噬发生和线粒体生成途径,改善糖尿病性肝脏病变。结论小檗碱具有改善糖尿病性肝脏病变的作用。 Objective To investigate the effects of berberine on diabetic liver disease and the related molecular mechanisms involving mitochondria.Methods Twenty-five db/db mice and 12 C57 mice were divided into three groups:normal group(WT),db/db group(db/db)and db/db+berberine group(db/db+BBR).The morphology of liver tissue was observed via hematoxylin and eosin(HE)staining.Biochemical kits were used to detect the levels of total cholesterol(TC),total triglyceride(TG),alanine transaminase(ALT),aspartate transaminase(AST),high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C).Western blotting was applied to detect the changes of mitochondria-associated proteins.Results Compared with the normal group,the levels of TG,TC and LDL-C in the db/db group were significantly increased,and the levels of HDL-C were decreased,and the levels of ALT and AST proteins were also remarkably enhanced.The liver function was significantly improved in the db/db+BBR group as compared with the db/db group(P<0.05).At the same time,berberine significantly ameliorated vacuolation in the liver tissues caused by diabetes.In addition,mitophagy and mitochondrial biogenesis were activated by berberine to ameliorate diabetes-induced liver disease.Conclusion Berberine can improve diabetes-induced liver disease.
