摘要
目的:探讨地西他滨(DAC)对人卵巢癌细胞株SKOV3的细胞凋亡和增殖等细胞生物学方面的影响.方法:采用不同浓度的地西他滨(10、100、 200 nmol/L)处理人卵巢癌细胞株SKOV3后,通过流式细胞仪检测地西他滨对人卵巢癌细胞株SKOV3的细胞凋亡的影响,细胞活性检测试剂盒(CCK-8)检测地西他滨、顺铂(Cisplatin)以及地西他滨联合顺铂对人卵巢癌细胞株SKOV3的增殖的影响.结果:浓度为100 nmol/L的地西他滨对人卵巢癌细胞株SKOV3的增殖抑制最明显,细胞增殖抑制率达61.31%,有统计学差异(P<0.05).地西他滨浓度超过100 nmol/L后对SKOV3的抑制率没有明显升高.低浓度地西他滨联合顺铂相比于单用地西他滨或者顺铂,SKOV3的细胞增殖抑制率升高,有统计学差异(P<0.05).浓度为100 nmol/L的地西他滨处理人卵巢癌细胞株SKOV3后细胞凋亡率升高,有统计学差异(P<0.05).结论:低浓度地西他滨促进人卵巢癌细胞株SKOV3的凋亡,抑制SKOV3的增殖,地西他滨可能通过增强铂类化疗药物的敏感性抑制肿瘤的发生发展.
Objective:To investigate the effects of decitabine on the cell apoptosis and cell proliferation of human ovarian cancer cell line SKOV3.Methods:The SKOV3 cells were treated with different concentrations(10,100,200 nmol/L)of decitabine.Flow cytometry was used to detect the effect of decitabine on cell apoptosis in SKOV3 cells.Cell counting kit-8 was used to detect the effect of decitabine,cisplatin and decitabine combined with cisplatin on cell proliferation in SKOV3 cells.Results:The inhibition effect decitabine at 100 nmol/L on the proliferation of SKOV3 cells was most obvious,and the growth inhibition rate was 61.31%(P<0.05).The inhibition rate was not significantly increased wtih decitabine over 100 nmol/L.Compared with decitabine or cisplatin alone,decitabine at low concertration combined with cisplatin caused a higher inhibition rate of cell proliforation(P<0.05).The apoptosis rate was increased in SKOV3 cells after the treatment with decitabine(P<0.05).Conclusion:Low dose decitabine promoted cell apoptosis and inhibied cell proliferation in SKOV3 cells.Low dose decitabine may prevent tumor development through enhancing the chemotherapy sensitivity of cisplatin.It provided a new idea for the treatment of drug-resistant ovarian cancer.
