炎症性肠病中MT、NF-κB及HIF-1α表达的相关性 被引量:4

Expression and significance of MT,NF-κB and HIF-1α in patients with inflammatory bowel disease

摘要 目的研究金属硫蛋白(MT)、核因子-κB(NF-κB)及缺氧诱导因子-1α(HIF-1α)在炎症性肠病患者的结肠组织及外周血中的表达及临床意义。方法收集2017-062018-06郑州大学第二附属医院住院的炎症性肠病患者结肠组织活检石蜡包埋标本及血清样本,其中UC患者30例,CD患者30例,30例结肠癌手术切除癌旁结肠组织作为组织对照,30例健康志愿者血清标本作为血清对照。分别采用免疫组织化学SABC检测结肠黏膜中MT和NF-κB的表达,ELISA检测外周血血清HIF-1α的含量。结果 MT、NF-κB在各组结肠组织中的表达差异均有统计学意义,HIF-1α在各组外周血血清中的表达差异有统计学意义(P <0. 01)。其中,MT的表达UC组及CD组低于对照组,NF-κB及HIF-1α的表达UC及CD组高于对照组。MT与NF-κB呈负相关(r=-0. 792,P <0. 01); MT与HIF-1α呈负相关(r=-0. 746,P <0. 01); NF-κB与HIF-1α呈正相关(r=0. 814,P <0. 01)。结论 MT表达与NF-κB及HIF-1α表达之间具有相关性,MT表达变化可能影响NF-κB及HIF-1α的表达参与炎症性肠病的发病过程。 Objective To investigate the expression and clinical significance of metallothionein(MT),nuclear factor-κB(NF-κB)and hypoxia-inducible factor-1α(HIF-1α)in colonic and peripheral blood of patients with inflammatory bowel disease.Methods Paraffin embedded specimens of colon tissue biopsy and serum samples in patients with inflammatory bowel disease hospitalized in the Second Affiliated Hospital of Zhengzhou University from June 2017 to June 2018 were collected from 30 cases of ulcerative colitis(UC),30 cases of Crohn';s disease(CD).The normal intestinal mucosa tissues from surgical specimens of 30 colorectal cancer patients were collected as tissue control,and blood samples from 30 healthy volunteers were collected as serum control.The expression of MT and NF-κB in colonic mucosa was detected by immunohistochemical SABC,and the content of HIF-1αin peripheral blood was detected by ELISA.Results The expression of MT and NF-κB in the colon tissue was statistically significant among different colon tissues.The expression of HIF-1αin the serum of peripheral blood was statistically among different groups(P<0.01).MT expression was lower in UC and CD groups than in control group.The expression of NF-κB and HIF-1αin UC and CD groups was higher than that in control group.MT was negatively correlated with NF-κB(r=-0.792,P<0.01)and HIF-1α(r=-0.746,P<0.01).NF-κB was positively correlated with HIF-1α(r=0.814,P<0.01).Conclusion There is a correlation between MT expression and the expression of NF-κB and HIF-1α.The changes of MT expression may affect the expression of NF-κB and HIF-1αin the pathogenesis of inflammatory bowel disease.
