单细胞转录组测序分析人类胚胎发育阻滞的潜在机制 被引量:1

摘要 目的探讨胚胎发育阻滞的分子机制,为提高辅助生殖成功率提供新思路。方法单细胞转录组测序技术研究发育阻滞胚胎(包括体外受精和胞质内单精子注射过程)及对照组的转录组情况,利用加权基因共表达网络分析(WGCNA)等生物信息学手段分析与胚胎阻滞相关的信号通路和关键基因。结果阻滞胚胎中与细胞分裂、细胞周期等相关的信号通路表达异常。与ICSI组相比,IVF组阻滞胚胎的细胞骨架、氧化磷酸化等通路表达异常。结论通过单细胞转录组测序技术分析阻滞胚胎转录组的结果说明细胞分裂、周期、代谢等通路可能影响胚胎正常发育,ICSI过程影响了线粒体功能、结构的变化。 Objective To explore the molecular mechanism of embryonic development retardation.Methods Single cell transcriptomic sequencing technology was used to study the transcriptome of the human arrested embryos(with IVF and ICSI procedures)and the normal development embryos.The signal pathways and key genes related to the embryonic retardation were analyzed by weighted gene co-expression network analysis(WGCNA)and other bioinformatics tools.Results The expression of signal pathway related to cell division and cell cycle was abnormal in arrested embryos.Also,the pathways of the cytoskeleton and oxidative phosphorylation were significantly differentially expressed in IVF embryos compared with ICSI embryos.Conclusion The study shows that signal pathways related to cell division and cell cycle,cytoskeleton and oxidative phosphorylation are abnormal in arrested embryos.
