Expression and significance of miR-1228-5p in serum and primary liver cancer tissues

目的:探讨mi R-1228-5p在原发性肝细胞癌组织和血清中的表达及意义。方法:采用mi RNA快速提取试剂盒分别抽取患者和健康体检者血清中的总mi RNA及患者肝癌组织和癌旁正常肝组织中的总mi RNA,采用实时荧光定量PCR检测总mi RNA中mi R-1228-5p的相对表达量;分析mi R-1228-5p的相对表达量与患者年龄、性别、家族史、肿瘤最大径和临床分期的相关性;此外,用ROC曲线下面积分析mi R-1228-5p在原发性肝细胞癌和健康人群中的诊断价值。结果:原发性肝细胞癌患者血清中mi R-1228-5P的表达量明显高于健康体检者(P<0.05),并且mi R-1228-5P在原发性肝细胞癌组织中的表达量明显高于其癌旁正常肝组织(P<0.05);mi R-1228-5p表达水平与患者的年龄、性别及家族史无关(P>0.05),而与患者的肿瘤最大径和临床分期有关(P<0.05);mi R-1228-5p鉴别原发性肝细胞癌与健康人群中的敏感度和特异度分别为71%和93%。结论:mi R-1228-5p在肝癌患者中的表达水平高于健康人群,且其在肝癌组织中的表达水平高于癌旁正常肝组织。mi R-1228-5p的表达水平与患者的肿瘤最大径和临床分期相关。此外,mi R-1228-5p具有诊断原发性肝细胞癌的潜在价值。

Objective:To investigate the expression and significance of miR-1228-5p in the serum of primary liver cancer tissues and patients.Methods:Total miRNAs in the serum of patients and healthy subjects,total miRNAs in liver cancer tissues and healthy liver tissues of patients were extracted using miRNA rapid extraction kit.Real-time quantitative PCR was used to detect the relative miR-1228-5p in total miRNAs.The amount of expression;and the correlation between the relative expression of miR-1228-5p and the patient's age,gender,family history,tumor maximum diameter and clinical stage;in addition,the area under the ROC curve was analyzed for miR-1228-5p in primary Diagnostic value in liver cancer and healthy people.Results:The expression of serum in patients with primary liver cancer was significantly higher than that in healthy subjects(P<0.05),and the expression of miR-1228-5P in the median value of hepatocarcinoma in patients with primary liver cancer was significantly higher than that in normal liver.Organization(P<0.05).The expression level of miR-1228-5p was not correlated with the age,gender and family history of the patients(P>0.05),but correlated with the tumor diameter and clinical stage of the patients(P<0.05).In addition,the sensitivity and specificity of miR-1228-5p in primary liver cancer and healthy people were 71%and 93%,respectively.Conclusion:The expression level of miR-1228-5p in liver cancer patients is higher than that in healthy people,and its expression level in liver cancer tissues is higher than that in normal liver tissues.The expression level of miR-1228-5p is related to the tumor's maximum diameter and clinical stage.In addition,miR-1228-5p has the potential to diagnose primary liver cancer.
