梯度有序支架材料诱导神经干细胞的定向迁移

Directional migration of neural stem cells induced by orderly gradient scaffolds

摘要 基质细胞衍生因子-1α(stromal-cell-derived factor-1α, SDF1α)在诱导神经干细胞(neural stem cells, NSCs)迁移中发挥重要作用.采用静电纺丝技术制备放射状梯度有序纤维支架材料负载胶原特异结合的SDF1α(collagen-binding domain SDF1α, CBD-SDF1α),针对神经干细胞及神经干细胞球(neural stem cell sphere, NSCS)迁移效果的影响进行了研究.实验结果表明:梯度支架可诱导神经干细胞以及神经干细胞球沿着有序纤维支架从CBD-SDF1α低浓度区域向高浓度区域迁移;神经干细胞球在CBD-SDF1α高浓度区域扩散出的神经干细胞数量和扩散面积比CBD-SDF1α低浓度区域显著增加;神经干细胞球沿着梯度有序纤维方向上呈现极化及定向延伸. Stromal-cell-derived factor-1α(SDF1α)plays an important role in inducing stem cell migration.Using a modified electrospinning technique,CBD-SDF1αembedded in radial orderly gradient fiber scaffolds to study migration of neural stem cells(NSCs)and neural stem cell spheres(NSCS)was prepared.The following results were obtained.1)Gradient scaffolds induced migration of NSCs and NSCS along the orderly fiber scaffolds from a low concentration area to a high concentration area of collagen-binding domain SDF1α(CBD-SDF1α).2)The number and diffusion area in NSCs derived from NSCS were increased significantly in a high concentration area than in a low concentration area of CBD-SDF1α.3)NSCS exhibited polarized morphology and extended directionally along aligned gradient fiber scaffolds.
