Expression and Clinical Significance of Ca MKⅡδ in Prostate Cancer

目的探讨钙/钙调素依赖蛋白激酶Ⅱδ(Ca MKⅡδ)基因在前列腺癌组织中的表达及意义。方法应用RT-PCR检测前列腺癌组织及其癌旁组织Ca MKⅡδmRNA表达情况,免疫组织化学法检测标本中Ca MKⅡδ蛋白表达及β-catenin表达情况,分析Ca MKⅡδ与前列腺癌PSA、Gleason评分、T分期、病理分期、淋巴结转移情况等临床病理参数的关系,并分析Ca MKⅡδ表达与β-catenin表达的相关性,探讨Ca MKⅡδ在前列腺癌组织中的表达及意义。结果 Ca MKⅡδmRNA及蛋白在人前列腺癌组织中过表达,且表达与前列腺癌病理分期、淋巴结转移、T分期显著相关,与PSA水平、Gleason评分无关,提示Ca MKⅡδ蛋白表达可能与前列腺癌侵袭、转移密切相关。前列腺癌组织中Ca MKⅡδ蛋白表达与β-catenin蛋白表达存在显著相关性。结论Ca MKⅡδ在人前列腺癌组织中过度表达,其表达与前列腺癌的外侵、淋巴结转移显著相关。

Objective To investigate the expression and clinical significance of CaMKⅡδin prostate cancer.Methods The expression of CaMKⅡδmRNA was detected in prostate cancer tissues and adjacent tissues with RT-PCR.Immuno-histochemistry was used to detect the expressions of CaMKⅡδprotein andβ-catenin and analyze the relationship between CaMKⅡδand prostate cancer PSA,Gleason score,T stage,pathological stage,and lymph node metastasis and the relationship between CaMKⅡδandβ-catenin expressions.Results CaMKⅡδmRNA and protein are overexpressed in human prostate cancer tissues,and the expression is positively correlated with pathological stage,lymph node metastasis,and T stage of prostate cancer,but is not related to PSA and Gleason score.These findings indicate that CaMKⅡδprotein expression is closely related to the invasion and metastasis of prostate cancer.The expression of CaMKⅡδprotein is significantly correlated with that ofβ-catenin protein in prostate cancer tissues.Conclusion CaMKⅡδis overexpressed in human prostate cancer tissues,and the expression is related to the invasion of prostate cancer and the metastasis of lymph node.
