摘要
【目的】旨在探讨饲粮添加鼠李糖乳酸杆菌GG(Lactobacillus rhamnosus GG,LGG)是否可以缓解轮状病毒(rotav-irus,RV)感染对断奶仔猪回肠黏膜屏障功能的影响。【方法】选取24头健康的21日龄断奶的"杜×长×大"仔猪,采用2×2因子试验设计,即LGG处理(添加或不添加109 CFU/kg LGG饲粮)和RV攻毒(灌服轮状病毒或无菌培养液),于试验第15天早上进行RV攻毒。试验期为19 d。【结果】RV攻毒诱导了断奶仔猪腹泻,显著提高了仔猪回肠黏膜中轮状病毒抗体和非结构蛋白4含量(P<0.05),显著提高了回肠黏膜的隐窝深度(P<0.05),显著降低了回肠黏膜的绒毛高度和绒毛高度/隐窝深度及ZO-1、occludin、mucin 1和mucin 2的表达(P<0.05)。饲粮中添加LGG显著提高了回肠黏膜的绒毛高度和绒毛高度/隐窝深度(P<0.05),有降低回肠黏膜隐窝深度的趋势(P=0.08),显著提高了回肠黏膜中ZO-1、occludin、mucin 1和mucin 2的表达(P<0.05);并降低了断奶仔猪的腹泻和回肠黏膜中非结构蛋白4含量(P<0.05),显著缓解了RV对回肠黏膜形态结构及ZO-1、occludin、mucin 1和mucin 2表达的影响(P<0.05)。【结论】饲粮添加LGG可以在一定程度上缓解RV诱导的断奶仔猪腹泻,而这一作用与其可改善仔猪回肠黏膜屏障功能有关。
【Objective】This trial was conducted to determine the effect of dietary Lactobacillus rhamnosus GG(LGG) supplementation on the ileal mucosa barrier function in the weaned pigs challenged by rotavirus(RV).【Method】Twenty-four healthy crossbred(Duroc×Landrace×Yorkshire) piglets weaned at 21 days of age were used in a 2×2 factorial design. The experimental factors included diets(control diet or supplementing GG diet) and RV challenge(oral gavage with RV or sterilized medium). On day 15,RV infusion was carried out. The experimental duration was 19 days.【Result】The results showed that RV challenge increased rate of diarrhea,enhanced the concentrations of rotavirus antibody and non-structural protein 4 in ileal mucosa(P<0.05),decreased villus height and villus height/crypt depth of ileal mucosa(P<0.05),increased crypt depth of ileal mucosa(P<0.05),and reduced the expressions of ZO-1,occludin,mucin 1 and 2 of ileal mucosa in weaned pigs(P<0.05). Dietary LGG supplementation reduced rate of diarrhea and rotavirus non-structural protein 4 level of ileal mucosa in the RV-infected pigs(P<0.05),and alleviated the effect of RV challenge on ileal mucosa morphology and expressions of ZO-1,occludin,mucin 1 and 2 of ileal mucosa(P<0.05).【Conclusion】These results suggested that dietary LGG supplementation,to some extents,relieved the RV-induced diarrhea,which was associated with the improvement of ileal mucosa barrier function in weaned pigs.
