CD151对结肠癌细胞WNT信号通路的影响机制分析 被引量:1

摘要 目的探讨CD151对结肠癌细胞WNT信号通路的影响机制。方法将20只BALB/c-nu/nu雄性裸鼠随机分为对照组和观察组,每组各10只,对照组裸鼠腋下注射结肠癌细胞HT29,观察组裸鼠腋下注射CD151基因敲除的结肠癌细胞CD151--HT29。待肿瘤模型制备成功后,取肿瘤组织,比较肿瘤组织中WNT信号通路相关基因及蛋白的表达变化。结果对照组肿瘤组织中存在明显的CD151、β-catenin和β-actin蛋白的表达,而观察组的肿瘤组织中并不存在CD151的表达。对照组β-catenin相对表达量为(0.18±0.02),与观察组的(0.13±0.01)比较,差异无统计学意义(P﹥0.05)。观察组裸鼠的肿瘤组织中Wnt1a蛋白、Oct4蛋白表达量均低于对照组,差异均有统计学意义(P﹤0.05)。且与对照组相比,观察组的β-catenin mRNA发生显著性下调。结论 CD151可能是WNT信号通路的上游基因,对WNT信号通路相关蛋白表达有调控作用。 Objective To explore the effect of CD151 on the WNT signaling pathway in colorectal cancer cells.Method The research included 20 BALB/c-nu/nu male nude mice which were randomized as control group and study group,with 10 in each.The control group was injected with colon cancer cells(HT29)subcutaneously in the armpit,while colon cancer cells with CD151 knocked out(CD151^--HT29)were injected in study group instead.After the tumor models were successfully prepared,tumor tissues from all mice were collected to compare the change of the expression of genes and proteins related to WNT signaling pathway.Result The expression of CD151,β-catenin andβ-actin in the tumors of the control group were readily noticeable,while there was no expression of CD151 observed in study group.The relative expression ofβ-catenin in control group was(0.18±0.02),which showed no significant difference compared with the(0.13±0.01)in study group(P>0.05).The expression of Wnt1a and Oct4 in the tumor tissues of the study group was significantly lower than that of control group(P<0.05).Theβ-catenin mRNA was significantly down-regulated in study group compared with control group.Conclusion CD151 may be the upstream gene of WNT signaling pathway,which regulates the expression of WNT related protein.
