摘要 目的:探讨低剂量镉(Cadmium,Cd)暴露对分化破骨细胞骨骼吸收活性的影响。方法:M-CSF (25 ng/mL)+RANKL (30 ng/mL)诱导RAW264. 7细胞向破骨细胞分化,同时加入不同浓度Cd。通过实时荧光定量PCR实验,TRAP染色实验和骨吸收活性分析实验研究Cd对破骨细胞分化与活化的影响。结果:低浓度Cd能够促进破骨细胞活化相关基因Cathepsin K mRNA和MMP-9 mRNA的表达,刺激TRAP阳性细胞的分化,并增强分化破骨细胞的骨吸收活性。结论:低浓度Cd能够促进破骨细胞活化,表现为骨吸收活性升高。 Objective:To investigate the effects of low dose cadmium(Cd)exposure on bone resorption activity of differentiated osteoclast.Methods:RAW264.7 cells were induced to differentiate into osteoclast by M-CSF(25 ng/mL)+RANKL(30 ng/mL),and various concentrations of Cd were added into the cells.The effects of Cd on the differentiation and activation of osteoclast were investigated by real-time quantitative PCR,TRAP staining and bone resorption assay.Results:Low concentrations of Cd could promote the expression levels of Cathepsin K mRNA and MMP-9 mRNA,stimulate the differentiation of TRAP-positive cells,and enhance the bone resorption activity of differentiated osteoclast.Conclusions:low concentrations of Cd can promote osteoclast activation,which presented that bone resorption activity is elevated.
