龙牙百合和兰州百合远缘杂交胚拯救及杂种苗快繁体系的建立 被引量:3

摘要 远缘杂交是培育食用百合新品种的重要途径,为克服龙牙百合和兰州百合远缘杂交受精后障碍,本研究采用胚珠培养的方法成功获得了杂种苗,并对其增殖技术进行了研究。结果表明:适宜胚拯救的配方为:1/2 MS+6-BA0. 5 mg·L^(-1)+NAA0. 1 mg·L^(-1),萌发率达66. 67%;适宜增殖的配方为:MS+6-BA0. 2 mg·L^(-1)+NAA0. 5 mg·L^(-1),增殖率达2. 09倍;适宜增重及生根的配方为:MS+PP_(333)3. 0 mg·L^(-1)+NAA0. 2 mg·L^(-1)+AC 3. 0 g·L^(-1)+糖80g·L^(-1),单个鳞球平均增重达151. 67 mg,生根率达100%。该研究对获得食用百合优良杂种后代具有一定的理论意义和实际应用价值。 Distant Crossing is an important method to breed edible lily cultivars.In order to obtain hybrids of distant crossing of Longya lily and Lanzhou lily,the best methods of embryo rescue and hybrid proliferation were studied.Main results were obtained as follows:The formula for embryo rescue was:1/2MS+6-BA0.5mg·L^-1+NAA0.1mg·L^-1,The germination rate of ovule was 66.67%;The basic medium of proliferation was:MS+6-BA0.2 mg·L^-1+NAA0.5mg·L^-1,and the proliferation rate is 2.09 times.the best medium for weight and rooting was MS+PP 333 3.00 mg·L^-1+NAA0.2mg·L^-1+AC 3.00 g·L^-1+sucrose 80g·L^-1,The weight of single bulblet increasing was 151.67 mg,The rate of rooting is 100%.This research could show much theoretic importance and practical application value to obtain the excellent hybrid progeny of edible lily.
