金银花、益母草和蒲公英提取液对胰脂肪酶活性的影响

The effect of Flos Lonicera,Leonurus japonicas and Dandelion extract solutions on Pancreatic Lipase activities

目的观察金银花、益母草和蒲公英提取液对胰脂肪酶活性的影响。方法水提法分别提取金银花、益母草和蒲公英单方提取液、提取液复配以及混方提取液,按照国家标准GB/T 23535—2009的方法观察其对胰脂肪酶活性的影响,荧光光谱法检测其对酶结构的影响。结果提取液对胰脂肪酶的抑制效果为蒲公英单方提取液>益母草单方提取液>金银花单方提取液>复方提取液>提取物混合液。荧光光谱分析发现,提取液的加入产生了荧光淬灭现象,最大发射波长发生红移,而且淬灭呈现规律性的变化。结论金银花、益母草和蒲公英提取液对脂肪酶活性有抑制作用,这种抑制作用是通过改变酶结构的方式实现的。

Objective To observe the effect of Flos Lonicera,Leonurus japonicas and Dandelion on pancreatic lipase activities. Methods The single,mixture and compound solutions from Flos Lonicera,Leonurus japonicas and Dandelion were extracted by water.The pancreatic lipase activities were analyzed by national standard GB/T 23535-2009,and the enzyme structure was tested by fluorescence spectrometry. Results The comparative inhibitory effects of extract solutions on pancreatic lipase activities were as follow: Dandelion single extraction> Leonurus japonicus single extract solution> Flos Lonicera single extract solution>mixture extract solution>compound extract solution.The results of fluorescence spectrum analysis showed that the fluorescence quenching phenomenon occurred when the extract solutions were added,and the maximun emission wavelength was redshifted.The results also showed that the quenching appeared regular changes. Conclusion The pancreatic lipase acitivities were inhibited,and the enzyme structure was also changed by the extract solutions from Flos Lonicera,Leonurus japonicas and Dandelion .
