大鼠急性脑梗死后肾功能的动态观测 被引量:2

The dynamic observation of renal function after acute cerebral infarction in rats

摘要 目的观察大鼠急性脑梗死后不同时间点大鼠肾脏形态及其功能的改变。方法将48只SD雄性大鼠运用随机数字表法分为假手术组、模型组,每组再随机分为1 d、3 d、7 d、14 d 4个亚组,每个亚组6只,采用改良线栓法建立右侧大脑中动脉闭塞模型,分别在1 d、3 d、7 d、14 d测两组大鼠血清肌酐(Cr)、尿素氮(BUN)、胱抑素C(Cys C)的含量,并取肾组织进行HE及Masson染色,光镜观察肾组织的病理形态学改变。结果大鼠脑梗死后模型组在3 d、7 d、14 d时血清Cr、BUN、Cys C浓度显著升高,与假手术组比较差异有统计学意义(P <0. 05),且Cr含量在第14天最高,BUN含量在第7天最高,Cys C含量在第3天时最高;光镜观察发现脑梗死大鼠的肾组织细胞在1 d、3 d时有轻微肿胀,至7 d时出现肾小管上皮细胞坏死、刷状缘消失,第14天时损伤持续加重。结论大鼠急性脑梗死后其肾脏结构和功能均受到一定损害。 Objective To observe the renal function and morphological changes after acute cerebral infarction in rats.Methods A total of 48 male SD rats were randomly divided into sham-operated group and model group.The right middle cerebral artery occlusion (MCAO) model was established by modified suture embolization method.Serum creatinine (Cr),Blood urea nitrogen (BUN),and cystatin C (Cys C) levels were measured at 1 d,3 d,7 d,and 14 d,respectively.The brain and kidney tissues were fixed,sectioned and stained with HE.The pathological changes of brain and kidney tissues were observed by light microscopy.Results The serum levels of Cr,BUN,and Cys C increased significantly at 3 d,7 d,and 14 d after cerebral infarction in rats,and there was a statistically significant difference from the sham-operated control group (P<0.05),and the Cr content was the highest level at the 14th day.BUN content was the highest at the 7th day,and Cys C content was the highest at the 3rd day.Light microscope observation showed that renal tissue cells of rats with cerebral infarction had slight swelling at 1 d and 3 d,and tubular necrosis and brushing appeared at 7 d.The margin disappeared and the damage continued to increase on the 14th day.Conclusion The renal function and renal structure were damaged after acute cerebral infarction in rats.
