微波提取红芪总多糖工艺及其抗氧化活性的研究 被引量:13

摘要 用微波提取红芪总多糖,采用DPPH、O_2^-·和ABTS三种自由基测定抗氧化活性,考察微波提取时间、微波功率、料液比对红芪总多糖得率及其抗氧化活性的影响。结果表明,红芪总多糖微波提取的最佳工艺为:以蒸馏水为提取溶剂,料液比1∶15 g/mL,微波功率231 W,提取15 min。此工艺条件下,多糖的得率为3.52%,DPPH清除率为81.4%。总多糖提取物具有较好体外抗氧化活性,高浓度的多糖抗氧化活性相当于0.5 mg/mL VC,总多糖对DPPH、O_2^-·和ABTS的IC50分别为3.777,3.727,4.423 mg/mL,且呈一定的量效关系。 Microwave extract total polysaccharides from Radix Hedysari were studied.Three methods,DPPH,O^-2·method and ABTS,were used to determine the antioxidant activity of the extracts.The effects of microwave extraction time,microwave power and material and liquid ratio,on the total polysaccharide yield and antioxidant activity of Radix Hedysari were investigated.Results showed that the optimum microwave extraction process of Radix Hedysari was as follows:distilled water was used as extraction solvent,solid-liquid ratio was 1∶15 g/mL,microwave power was 231 W,15 min was extracted,the yield of polysaccharide was 3.52%,and the clearance rate of DPPH was 81.4%.The total polysaccharide extract in this process had better antioxidant activity in vitro,the high concentration of polysaccharide antioxidant activity was equivalent to 0.5 mg/mL VC,and the total polysaccharide was 3.777,3.727,4.423 mg/mL to the IC 50 of DPPH,O^-2·method and ABTS,respectively.The antioxidant activity of the extract is high and has a certain dose effect relationship.
