摘要 [目的]建立同时测定霍山黄大茶中5种儿茶素类成分及咖啡因的UPLC定量分析方法。[方法]采用梯度洗脱法同时测定EC、ECG、EGC、EGCG、GC、caffeine,选用Waters BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm),流动相为乙腈-甲酸-水梯度洗脱,体积流量为0.2 mL/min,柱温设为30℃,检测波长210 nm。[结果]测定了霍山黄大茶中儿茶素类和咖啡因的含量,EC、ECG、caffeine、EGC、EGCG和GC分别在相应范围内线性关系良好(r>0.999 5),平均回收率分别为98.82%、97.30%、96.96%、96.59%、98.89%和100.11%,RSD为1.0%~1.8%。[结论]该方法快速简捷、灵敏度高、重现性好,可用做霍山黄大茶质量分析方法。 [Objection]The research aimed to establish an UPLC method for determination of five elected catechins and caffeine in Huoshan Yellow Big Tea.[Method]The extracts of the tea by ultrasonic extraction was analyzed on a Waters BEH C 18 column (2.1 mm × 100 mm,1.7 μm) with a mobile phase of acetonitrile-water (containing 0.5% formic acid) for gradient elution at 210 nm,flow rate was set at 0.2 mL/min.[Result]The content of catechins and caffeine in Huoshan Yellow Big Tea was determined.EC,ECG,caffeine,EGC,EGCG and GC had good linear relations in the corresponding range (r>0.999 5),and the average recoveries were 98.82%,97.30%,96.96%,96.59%,98.89%,100.11%,respectively.RSD was 1.0%-1.8%.[Conclusion]The method is quick and simple,has high sensitivity and good reproducibility,and can be used as a quality analysis method of Huoshan Yellow Big Tea.
