芦根提取物与苇根提取物HPLC特征图谱的比较 被引量:6

摘要 目的比较芦根提取物与苇根提取物HPLC特征图谱的差异,用以鉴别芦根提取物及其伪品。方法采用ZORBAX SB C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.5%KH_2PO_4(pH=2.15),梯度洗脱;检测波长为275 nm;柱温为30℃;流速为1.0 mL/min。采用国家药典委员会"中药色谱指纹图谱相似度评价系统软件(2004A版)"生成对照图谱。结果 10批芦根提取物特征图谱共标示出包括对香豆酸、阿魏酸在内的7个特征峰。10批苇根提取物特征图谱共标示出13个特征峰,其中包括芦根提取物的7个特征峰,且比芦根提取物多了6个易于辨认的黄酮特征峰。与对照图谱比较,10批芦根提取物、苇根提取物的相似度分别为0.935~0.999、0.807~0.973,芦根提取物与苇根提取物共有模式之间的相似度为0.875。结论本方法准确可靠、重复性好,可作为芦根提取物鉴别和质量控制的评价依据。 Objective To compare the difference of HPLC characteristic spectra of Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract, so as to identify the Phragmitis Rhizoma extract and its adulterants. Methods HPLC was performed on ZORBAX SB C 18 column(250 mm×4.6 mm,5 μm), the acetonitrile-0.5% potassium dihydrogen phosphate aqueous solution (pH2.15) was employed as a mobile phase at gradient elution, detection wavelength was 275 nm;column temperature was 30℃, and flow rate was 1.0 mL/min. The contrast spectrum was generated with Traditional Chinese medicine (TCM) chromatographic fingerprint similarity evaluation software system (2004A version) by State Pharmacopoeia Commission. Results Seven common peaks were separated from 10 batches of Phragmitis Rhizoma extract, including p -coumaric acid and ferulic acid. Thirteen common peaks were separated from 10 batches of Zizaniae caduciflorae Rhizoma extract, which included 7 same common peaks of Phragmitis Rhizoma extract and six common peaks (peak w1- peak w6) of flavonoids easily recognized. The similarities of 10 batches of Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract were 0.935-0.999,0.807-0.973 respectively, and the similarity of the mutual modes between Phragmitis Rhizoma extract and Zizaniae caduciflorae Rhizoma extract was 0.875. Conclusion The method is accurate, reliable and reproducible, which can be used for the identification and quality evaluation of Phragmitis Rhizoma extract.
