摘要
目的研究丹参提取物对庆大霉素导致的HEI-OC1细胞和斑马鱼毛细胞损伤的影响,并探讨丹酚酸B对庆大霉素耳毒性的保护作用。方法先用丹参提取物或丹酚酸B对HEI-OC1细胞和野生AB系斑马鱼进行预处理,再利用庆大霉素造模,采用MTT法检测HEI-OC1的细胞存活率,使用DASPEI对斑马鱼毛细胞染色并进行半定量分析。结果 HEI-OC1细胞损伤模型中,丹参提取物组及丹酚酸B组的细胞存活率均高于庆大霉素造模组(P<0.01);斑马鱼毛细胞损伤模型中,丹参提取物组的荧光强度值高于庆大霉素造模组(P<0.01),而丹酚酸B组差异无统计学意义。结论丹参提取物对庆大霉素引起的耳毒性有一定保护作用,丹酚酸B的稳定性经改善后有望成为潜在的耳毒性保护药物。
Objective To investigate the protective effects of Salvia miltiorrhiza extract and salvianolic acid B (Sal B) against drug-induced hearing loss (DIHL). Methods HEI-OC1 cells and zebrafish were exposed to gentamicin after pretreated by Salvia miltiorrhiza extract or Sal B. The viability of HEI-OC1 cells was analyzed by MTT assay and the damage of the zebrafish hair cells was detected using DASPEI staining. Results In HEI-OC1 cell experiment,the cell viability of Salvia miltiorrhiza extract or Sal B group was higher than that of gentamicin group ( P< 0.01). In the zebrafish hair cell experiment,the hair cell fluorescence intensity in Salvia miltiorrhiza extract group was more than that of gentamicin group ( P< 0.01), but Sal B group had no significant difference. Conclusion Salvia miltiorrhiza extract has protective effects against DIHL,and Sal B will be a potential ototoxic therapeutic agent after improving its stability.
