四妙勇安汤对大鼠股骨干骨折术后抗凝作用的实验研究

Experimental Study on the Anticoagulation Effect of Simiao Yongan Decoction on Femoral Shaft Fracture in Rats

目的探讨四妙勇安汤对大鼠股骨干骨折术后抗凝的效果研究。方法将造模后的57只老鼠分为2组,实验组29只大鼠采用灌胃针从嘴角注射四妙勇安汤,2次/d,2 mL/只,连续1周;对照组28只大鼠的颈后缘皮肤提起后,注射器沿纵轴方向剌入皮肤注射低分子肝素钠注射液,2次/d,1 mL/只,连续1周。于造模后1 d、3 d、7 d采用眼眶取血,监测凝血指标D-二聚体(D-D)、凝血酶原时间(PT)、抗凝血酶Ⅲ(AT-Ⅲ)水平。结果造模后第1天2组大鼠D-二聚体水平差异无统计学意义(P>0.05),造模后第3天水平升高,且造模后第7天水平显著降低,实验组水平均显著低于对照组,2组比较差异有统计学意义(P<0.05)。造模后第1天2组大鼠PT、AT-Ⅲ水平差异无统计学意义(P>0.05),造模后第3天水平降低,且造模后第7天水平显著升高,实验组水平均显著高于对照组,2组比较有显著性差异(P<0.05)。结论四妙勇安汤治疗大鼠股骨干骨折术后抗凝效果明显,能够显著降低D-二聚体水平,延长凝血酶原时间,升高抗凝血酶Ⅲ水平,有效抑制骨折术后静脉血栓及凝血,为下步推广运用于临床奠定基础。

Objective To investigate the anticoagulation effect of Simiao Yongan decoction on postoperative femoral shaft fracture in rats.Methods After modeling,57 rats were divided into two groups.In the experimental group,29 rats were injected with Simiao Yongan decoction from the corner of the mouth with an intragastric needle,2 times/day,2 ml/only for 1 week,the back of the neck of 28 rats in the control group.After the skin was lifted,the syringe was injected into the skin along the longitudinal axis to inject low molecular weight heparin sodium injection twice a day/day,1 ml/day for 1 week.Blood was taken from the eyelids 1 day,3 days,and 7 days after modeling,and the blood coagulation indexes D-D(D-D),prothrombin time(PT),and antithrombin III(AT-III)levels were monitored.Results There was no significant difference in the level of D-dimer between the two groups on the first day after model establishment(P>0.05),the level was increased on the third day after modeling,and the level was significantly decreased on the 7th day after modeling.Both were significantly lower than the control group,and there was a significant difference between the two groups(P<0.05).There was no significant difference in the levels of PT and AT-III between the two groups on the first day after model establishment(P>0.05).The level was decreased on the third day after modeling,and the level was significantly increased on the 7th day after modeling.They were significantly higher than the control group,and there was a significant difference between the two groups(P<0.05).Conclusion Simiao Yongan decoction has obvious anticoagulant effect after treatment of femoral shaft fracture in rats,which can significantly reduce D-dimer level,prolong prothrombin time,increase antithrombin III level,and effectively inhibit postoperative fracture.Venous thrombosis and coagulation are the basis for the next step to promote clinical use.
