摘要
体内药物分析实验为本校新开实验,建立可靠的体内药物分析方法尤其重要。通过对血清中对乙酰氨基酚分析方法进行探讨,使其适用于体内药物分析的学生实验。分析方法探讨主要包括样品前处理方法和高效液相色谱法色谱条件两方面,寻找得到最优分析方法,并对此方法进行方法学验证。结果表明:最优色谱条件为以茶碱为内标,A:甲醇-乙腈(5∶3),B:0.3%磷酸水溶液,A∶B=15∶85为流动相,检测波长254nm;最佳样品处理方法为沉淀蛋白法和液液萃取法,且方法学验证符合要求,可以用于体内药物分析学生实验。
The biopharmaceutical analysis experiment was a new experiment for our school, so it was especially important to establish a reliable analysis method. The aim of this study was to discuss the analysis method for the determination of acetaminophen in serum, making it suitable for student experiment of biopharmaceutical analysis. Analysis method mainly includes the sample pretreatment methods and HPLC chromatographic conditions, looking for ways to get the optimal analysis method, and validation method was used as described. The results show that the optimum chromatographic condition was theophylline as the internal standard, A: methanol-acetonitrile (5 ∶3),B: 0.3% phosphoric solution, A ∶B=15 ∶85 as the mobile phase and 254nm as the UV detective wavelength;the best sample processing method are the precipitation protein method and liquid-liquid extraction method, and the methodological validation meets the requirements, which can be used for student experiment of biopharmaceutical analysis.
