摘要
以刺激RAW264.7细胞释放NO活性模型为指导,通过乙醇沉淀和DEAE阴离子交换柱层析,从紫芝子实体中分离得到均一多糖GS30S1-20E,采用红外光谱、核磁共振、气相色谱-质谱法和高效液相等现代仪器,研究了GS30S1-20E的结构特性。结果表明:GS30S1-20E是一种β-D-葡聚糖,其重均相对分子质量为2.418×10~6,数均相对分子质量为1.668×10~6,多分散指数为1.450,特性粘度为927.49 mL/g,以β-(1,3)-糖苷键为主链和β-(1,6)-糖苷键为支链,主链与支链上糖残基的摩尔比为3∶1,具有刺激巨噬细胞释放NO的活性。
Crude polysaccharide GS30 precipitated from hot water extracts of the fruiting bodies of Ganoderma sinense were separated by DEAE-column chromatography,and then treated with ethanol precipitation to get a purified polysaccharide fraction GS30S1-20E. Modern analytical techniques,such as IR,NMR,GC/MS and HPSEC were conducted to analyze the structural characteristics of GS30S1-20E. Results showed that GS30S1-20E was a β-D-glucan with a high weight average molecular mass of 2.418×10^6 and number average molecular weight of 1.668×106,polydispersity of 1.450 and large viscosity of 927.49 mL/g. GS30S1-20E contains a backbone of(1→3)-linked-β-D-glucopyranosyl residues,bearing branches at the O-6 position of every three sugar residues along the backbone. An in vitro immunomodulating activity assay revealed that GS30S1-20E could significantly increase the release of NO from RAW 264.7 cells.
