摘要
通过对墨兰新品种‘梦之兰’的组织培养技术研究,探讨了不同激素浓度对‘梦之兰’的诱导启动、继代增殖、生根壮苗培养和不同基质配比对移栽成活率的影响,获得‘梦之兰’诱导启动最佳培养基为改良MS+6-BA1.5 mg·L^(-1)+KT0.6 mg·L^(-1)、最佳继代增殖培养基为改良MS+6-BA1.5 mg·L^(-1)+KT0.4 mg·L^(-1)+NAA0.6 mg·L^(-1)、最佳生根培养基为1/2改良MS+IBA1.0 mg·L^(-1)、最佳移栽基质为松树皮:珍珠岩=1∶1。
Through the study on the tissue culture technology of the new Cymbidium sinense variety ‘Mengzhilan’, We explored the effects of different hormone concentrations on the induction initiation, subculture, rooting and seedling culture and different matrix ratios of ‘Mengzhilan’ on the survival rate of transplanting. The results showed that the optimal medium for the induction to ‘Mengzhilan’ was Modified MS+6-BA1.5 mg·L -1 +KT0.6 mg·L -1 , and the best subculture medium was Modified MS+6-BA1.5 mg·L -1 +KT0.4 mg·L -1 +NAA0.6 mg·L -1 , the best rooting medium is 1/2 Modified MS+IBA1.0 mg·L -1 , The best transplanting substrate is pine bark∶perlite=1∶1.
