摘要
目的探讨榄香烯体外逆转肝癌HepG2细胞株耐索拉非尼的作用及其机制。方法购置肝癌HepG2细胞系,经过索拉非尼处理,获得索拉非尼肝癌耐药细胞;使用随机数字表,将获得的索拉非尼细胞株分为榄香烯组、索拉非尼组、榄香烯+索拉非尼组及阴性对照组,通过MTT实验、Western Blot检测等分析各组治疗效果。结果榄香烯+索拉非尼组细胞12 h、24 h抑制率,均高于榄香烯组、索拉非尼组及阴性对照组(P<0.05);榄香烯组细胞12 h、24 h抑制率,均高于索拉非尼组及阴性对照组(P<0.05);索拉非尼组细胞12 h、24 h抑制率,高于阴性对照组(P<0.05);榄香烯+索拉非尼组P21蛋白水平,高于榄香烯组、索拉非尼组及阴性对照组(P<0.05);榄香烯组P21蛋白水平,高于索拉非尼组及阴性对照组(P<0.05);索拉非尼组P21蛋白水平,高于阴性对照组(P<0.05);榄香烯+索拉非尼组Raf激酶水平,低于榄香烯组、索拉非尼组及阴性对照组(P<0.05);榄香烯组Raf激酶水平,低于索拉非尼组及阴性对照组(P<0.05);索拉非尼组Raf激酶水平,低于阴性对照组(P<0.05)。结论榄香烯能逆转肝癌细胞对索拉非尼的获得性耐药,且协同索拉非尼能发挥联合抗肿瘤作用,能为临床肝癌治疗提供新的思路与方法。
Objective To investigate the effect and mechanism of elemene on reversing the resistance of sorafenib in hepatocellular carcinoma (HCC) HepG2 cell line. Methods Hepatocellular carcinoma HepG2 cell line was purchased and treated with sorafenib to obtain sorafenib-resistant liver cancer-resistant cells. The obtained sorafenib cell line was divided into elemene group, sorafenib group, elemene+sorafenib group and negative control group with a random number table. All the group were analyzed by MTT assay and Western Blot for the results of the experiment. Results At 12 h and 24 h, the inhibition rates of the elemene+sorafenib group were higher than those of the elemene group, the sorafenib group and the negative control group (P<0.05). The inhibition rates of the elemene group was higher than that of the sorafenib group and the negative control group (P<0.05). The inhibition rate of the sorafenib group was higher than that of the negative control group (P<0.05). The level of P21 protein in the elemene+sorafenib group was higher than that in the elemene group, the sorafenib group and the negative control group (P<0.05). The P21 protein level in the elemene group was higher than that in the sorafenib group and the negative control group (P<0.05). The level of P21 protein in sorafenib group was higher than that in negative control group (P<0.05). The level of Raf kinase in the elemene+sorafenib group was lower than that in the elemene group and the sorafenib group and the negative control group (P<0.05). The level of Raf kinase in the elemene group was lower than that in the sorafenib group and the negative control group (P<0.05). The level of Raf kinase in the sorafenib group was lower than that in the negative control group (P<0.05). Conclusion Elemene can reverse the acquired resistance of sorafenib in HepG2 cell lines, and synergistic effects were seen with elemene and sorafenib, which can provide new ideas and methods for HCC treatment.
