摘要
目的:探讨桦褐孔菌乙酸乙酯提取物对食管癌细胞EC-109、EC-9706增殖、凋亡和细胞周期等表型的影响。方法:用桦褐孔菌乙酸乙酯提取物处理食管癌细胞株EC-109、EC-9706,采用MTT法、流式细胞术、平板克隆实验分析桦褐孔菌乙酸乙酯提取物对食管癌细胞增殖、凋亡和细胞周期的影响。结果:桦褐孔菌乙酸乙酯提取物可显著抑制食管癌细胞的增殖,并降低肿瘤细胞的克隆形成能力。流式细胞术显示,桦褐孔菌提取物可将食管癌细胞周期阻滞在G1期,并促进肿瘤细胞的凋亡。结论:桦褐孔菌乙酸乙酯提取物可抑制人食管癌细胞株的增殖和克隆形成,阻滞细胞周期并诱导肿瘤细胞凋亡,具有一定的抗食管癌活性。
Objective: To investigate the effects of ethyl acetate extract of Inonotus obliquus on proliferation,apoptosis and cell cycle phenotypes of esophageal cancer cells EC-109 and EC-9706. Methods: Ethyl acetate extract of I.obliquus was used to treat esophageal cancer cells EC-109 and EC-9706. MTT,flow cytometry and plate cloning assay were used to analyze the effects of ethyl acetate extract of Lobliquus on proliferation, apoptosis and cell cycle of esophageal cancer cells. Results: Ethyl acetate extract of I.obliquus could significantly inhibit the proliferation of esophageal cancer cells and reduce the cloning ability of cancer cells. Flow cytometry showed that the extract of Lobliquus could block the cell cycle of esophageal cancer in G1 phase and promote the apoptosis of cancer cells. Conclusion: Ethyl acetate extract of I.obliquus can inhibit the proliferation and cloning ability of human esophageal cancer cell lines, and block cell cycle and induce apoptosis of tumor cells, and has certain anti-esophageal cancer activity.
