摘要
《食品生物化学》实验实训"氨基酸的纸色谱"是常见氨基酸分析方法,为改善实验方法,提高教学质量,本文对氨基酸纸色谱层析法的固定相、展开剂、点样方式和显色方式进行优化和改进,经试验确定,以薄层板为固定相,V(正丁醇):V(甲酸):V(水)=15∶3∶2为流动相,移液枪点样,茚三酮加入展开剂为显色方式,达到更好的分离效果。
Paper chromatography is a common amino acid analysis method in the experiment of “food biochemistry”.In order to improve the quality of teaching,this paper optimizes and improves the fixed phase,the expansion agent,the point pattern and the color rendering method of the amino acid chromatography.Taking thin layer plate as stationary phase,V (n-butanol)∶ V (formic acid)∶ V (water)=15∶3∶2 as mobile phase,liquid shift gun point sample,and three branch of acetone add developer as color developing mode,so as to achieve the expected separation effect.
