摘要 目的:探讨EDTAK2、肝素钠、枸橼酸钠三种不同抗凝剂对流式细胞术检测二磷酸腺苷(ADP)介导的血小板膜糖蛋白CD62p表达的影响。方法:使用不同抗凝剂,采用流式细胞术检测ADP诱导大鼠外周全血中CD62p表达率。结果:枸橼酸钠对ADP诱导CD62p表达影响较小,EDTAK2明显抑制ADP介导的CD62p的表达,而肝素钠能明显促进CD62p的表达,同时使全血中CD62p自身表达偏高。结论:枸橼酸钠为流式细胞仪检测血小板活化的最佳抗凝剂,浓度3. 2%(含2个结晶水的枸橼酸钠)和3. 8%(含5个结晶水的枸橼酸钠)是其抗凝最佳方案。 Objective:To determine the adenosine diphosphate(ADP)induced expression of platelet membrane glycoprotein CD62p in different anticoagulants treated rats by flow cytometry.Methods:ADP-induced CD62p expression in peripheral blood of three different anticoagulants(EDTAK2,heparin sodium and sodium citrate)treated rats was detected by flow cytometry.Results:Sodium citrate had little effect on ADP-induced CD62p expression.EDTAK2 significantly inhibited the expression of ADP-induced CD62p,while heparin sodium significantly promoted the expression of CD62p and increased rat whole blood CD62p expression level.Conclusion:Sodium citrate is the best anticoagulant for the detection of platelet activation by flow cytometry,and the best anticoagulation concentrations of sodium citrate for detecting CD62p expression by flow cytometry are 3.2%(sodium citrate containing 2 crystalline water)and 3.8%(sodium citrate containing 2 crystalline water),respectively.
