电针预处理对心肌缺血再灌注无复流大鼠NF-kB p65蛋白及其磷酸化水平的影响 被引量:7

The Effect of Electroacupuncture Pretreatment on NF-kB p65 and Its Phosphorylation Levels in Rats with Myocardial No-reflow After Ischemia-reperfusion

摘要 目的观察电针预处理对大鼠心肌缺血再灌注无复流现象及核转录因子-?B(NF-?B) p65蛋白及其磷酸化水平的影响。方法 48只雄性SD大鼠随机分为假手术组、模型组、电针预处理组、阿托伐他汀预处理组。除假手术组外,其余各组大鼠均采用结扎左冠状动脉45 min后再灌注120 min的方法复制急性心肌缺血再灌注无复流的模型。观察各组大鼠心肌梗死、缺血及无复流的范围,酶联免疫吸附法(ELISA)测定血清炎性因子白细胞介素-10(IL-10)、白细胞介素-6(IL-6)和肿瘤坏死因子(TNF-?)的水平,蛋白质免疫印迹法(Western-blot)测定心肌组织中NF-?B p65及其磷酸化水平的表达。结果与模型组比较,电针预处理组和阿托伐他汀预处理组心肌梗死范围及无复流范围明显减小,电针预处理能显著升高心肌梗死大鼠血清IL-10水平,降低血清IL-6及TNF-?水平,下调心肌组织中NF-?B p65及phospho-NF-?B p65蛋白的表达,差异均有统计学意义(P<0.01,P<0.05)。结论电针预处理能够显著降低心肌组织中NF-?B p65及其磷酸化水平的表达,从而改善心肌缺血再灌注后的无复流现象。 Objective To observe the effect of electroacupuncture pretreatment on myocardial no-reflow after ischemia-reperfusion, NF-κB p65 and its phosphorylation levels in rats. Method Forty-eight male SD rats were randomized to sham operation, model, electroacupuncture pretreatment and atorvastatin pretreatment groups. A model of myocardial no-reflow after acute ischemia-reperfusion was made in all the groups except the sham operation group. The model was made by left coronary artery ligation for 45 min and subsequent reperfusion for 120 min. The extents of myocardial infarct, ischemia and no-reflow were assessed in every group of rats. Serum inflammatory factors interleukin-10 (IL-10), interleukin-6 (IL-6) and tumor necrosis factor (TNF-α) levels were measured by enzyme-linked immunosorbent assay (ELISA). Myocardial expressions of NF-κB p65 and phospho-NF-κB p65 were determined by Western blot. Result The extents of myocardial infarct and no-reflow were significantly reduced in the electroacupuncture pretreatment and atorvastatin pretr eatment groups compared with the model group. Electroacupuncture pretreatment could significantly raise serum IL-10 levels, reduce serum IL-6 and TNF-α levels and down-regulate myocardial expressions of NF-κB p65 and phospho-NF-κB p65 with statistically significant differences (P<0.01, P<0.05). Conclusion Electroacupuncture pretreatment can significantly decrease myocardial expressions of NF-κB p65 and phospho-NF-κB p65 to reduce myocardial no-reflow after ischemia-reperfusion.
