摘要
目的考察苏合香对大鼠心肌缺血的影响。方法冠状动脉结扎法制备大鼠心肌缺血模型。84只大鼠随机分为假手术组、模型组、溶剂组(5%吐温)、阳性药组(硝酸甘油溶液)及苏合香低、中、高剂量组(0.167、0.333、0.667 g/kg),每组12只。然后记录ST段、左心室终末舒张压(LVEDP)、左心室平均压(LVAP),TTC染色检测心肌梗死率,HE染色观察心肌组织,比色法检测血清肌酸激酶同工酶(CK-MB)活性,微板法检测血清乳酸脱氢酶(LDH)、谷草转氨酶(AST)活性。结果与溶剂组比较,苏合香中、高剂量组ST段显著升高(P<0.05,P<0.01),低、中剂量组LVEDP、LVAP及低剂量组心肌梗死率显著降低(P<0.05,P<0.01),各剂量组心肌损伤显著改善(P<0.05,P<0.01),CK-MB、LDH、AST活性显著下降(P<0.05,P<0.01)。结论苏合香有一定抗心肌缺血作用,并与其剂量呈负相关。
AIM To investigate the effects of Styrax on myocardial ischemia in rats. METHODS Myocardial ischemia rat model was induced by coronary artery ligation method. Eighty-four rats were randomly divided into sham operation group, model group, solvent group (5% Tween), positive drug group (nitroglycerin solution) and low-dose, medium-dose, high-dose Styrax groups (0.167, 0.333, 0.667 g/ kg), 12 rats in each group. Subsequently, ST segment, left ventricular end-diastolic pressure (LVEDP), left ventricular average pressure (LVAP) were recorded. TTC staining and HE staining were applied to detecting myocardial infarction rate and observing myocardial tissues, respectively. Colorimetry method and microplate method were adopted in the serum activity detection of creatine kinase isoenzyme ( CK-MB) and lactate dehydrogenase ( LDH), aspartate aminotransferase (AST), respectively. RESULTS Compared with the solvent group, the medium-dose, high-dose Styrax groups demonstrated significant elevated ST segment (P<0.05, P<0.01);the low-dose, medium-dose groups markedly decreased LVEDP, LVAP, and the low-dose group significantly reduced myocardial infarction rate (P<0.05, P< 0.01);the various dose groups exhibited obviously improved myocardial injury (P<0.05, P<0.01) and significantly reduced CK-MB, LDH, AST activities (P<0.05, P<0.01). CONCLUSION Styrax exhibits a certain anti-myocardial ischemia effect, which has a negative correlation with its dose.
