摘要 目的研究KIF18A在肺腺癌中的诊断和预测价值。方法通过对癌症基因组图谱(TCGA)数据库和郑州大学第一附属医院临床标本的分析,探讨KIF18A表达在预测肺腺癌患者预后中的作用。采用Kaplan-Meier分析研究KIF18A的表达水平与肺腺癌患者的生存时间之间的关系。采用多变量Cox回归模型研究KIF18A的表达水平能否独立判断肺腺癌患者的预后。结果通过对TCGA数据库的分析和临床组织标本的检验显示,KIF18A在肺腺癌中的表达量比在正常肺组织中高。Log-rank生存分析显示,KIF18A表达量高的患者总体存活率较低。KIF18A的高表达与患者的性别、临床分期、肿瘤分期和远处转移相关。多变量Cox回归模型分析显示,KIF18A的表达水平是肺腺癌患者总体生存状态的独立预测因子。结论 KIF18A在人肺腺癌中高表达,并且其表达水平是肺腺癌患者总体生存状态的独立预测因子。 Objective To analyze the diagnostic and predictive value of KIF18A in lung adenocarcinoma. Methods Based on the analysis of the Cancer Genome Atlas (TCGA) database and clinical specimens from the First Affiliated Hospital of Zhengzhou University,the role of KIF18A expression in forecasting the lung adenocarcinoma outcome was investigated. Kaplan Meier analysis was used to calculate the relation between the level of KIF18A expression and the over survival time of patients in lung adenocarcinoma. Multivariate Cox regression model was used to study whether the expression level of KIF18A could independently predict the prognosis of patients with lung adenocarcinoma. Results The TCGA and clinical data predicted that the gene expression levels of KIF18A were overexpressed in lung adenocarcinoma. Log-rank analysis of survival status revealed that patients with increased levels of KIF18A had a lower overall survival rate. In addition,increased KIF18A expression correlated with gender,clinical staging,tumor staging and distant metastasis. Multivariate Cox regression model showed that KIF18A mRNA level was an independent predictor for overall survival status of lung adenocarcinoma patients. Conclusion KIF18A is overexpressed in lung adenocarcinoma,and it is an independent predictor for overall survival status of lung adenocarcinoma patients.
