毛蕊花苷对大鼠递增大强度运动下骨骼肌损伤的保护作用研究 被引量:3

摘要 目的:研究毛蕊花苷对递增大强度运动大鼠骨骼肌损伤的保护作用。方法:SD大鼠随机分为正常对照组(NC)、单纯运动组(SE)、运动+毛蕊花苷低剂量组(EVL)、运动+毛蕊花苷中剂量组(EVM)、运动+毛蕊花苷高剂量组(EVH)。采用递增大强度跑台运动疲劳模型。实验末,取血和骨骼肌检测各组大鼠血清CK酶活性和骨骼肌氧化应激指标及HSP70蛋白表达量,并用光学显微镜观察骨骼肌微细结构改变。结果:1)与SE组相比,毛蕊花苷各剂量组明显减少了运动引起的血清CK活性的上升(P<0.05或P<0.01),减轻了运动所致的骨骼肌微细结构的异常,且EVH组的效果要好于EVL组和EVM组。2)与SE组相比,毛蕊花苷各剂量组显著抑制了运动导致的大鼠骨骼肌的氧化应激水平(P<0.05或P<0.01),其中EVH组抗氧化损伤的效果要好于EVL组和EVM组。3)与SE组相比,毛蕊花苷剂量组进一步诱导了运动引起的HSP70的高表达(P<0.05或P<0.01),其中以EVH组的蛋白表达增加更为显著(P<0.05)。结论:毛蕊花苷对运动大鼠骨骼肌损伤有良好的改善作用,显示出毛蕊花苷有明显抗骨骼肌氧化应激和减轻运动性骨骼肌微细结构损伤的保护作用,其中高剂量补充组的改善效果要优于其他剂量组,这可能与其能更好地诱导运动大鼠骨骼肌内源性保护蛋白(HSP70)表达以发挥HSP70保护效应有关。 Objective:To investigate protective effect of verbascoside on skeletal muscle injury induced exercise in trained rats.Methods:Sprague-Dawley(SD)rats were randomly divided into 5 groups:normal group(NC),simple exercise group(SE),exercise+verbascoside low dose group(EVL),exercise+verbascoside middle dose group(EVM)andexercise+verbascoside high dose group(EVH)with the mode of high-intensity increasing treadmill exercise fatigue.The serum CK was measured from the blood samples,and oxidative stress indexs and HSP70 protein expression were measured from skeletal muscle.And light microscope was employed to observe morphology changes of skeletal muscle in the experiment end.Results:1)Compared to SE group,verbascoside treatment declined the leakage of creatine kinase induced exercise(P<0.05 or P<0.01)and skeletal muscle ultrastructural malformation.Among this,the improvement effect of EVH group was better than the two groups.2)Compared to SE group,verbascoside treatment evidently alleviated oxidative stress by inhibiting lipiding effect of peroxidation(P<0.05 or P<0.01),and the improvement effect of EVH group was better than the two groups.3)Compared to SE group,verbascoside treatment induced significantly the high HSP70 expression in the exercise rats(P<0.05 or P<0.01).Conclusion:Verbascoside exhibited protective role in exercise-induced injuries of skeletal muscle,and the improvement effect of high dose supplement was better than other supplement groups,which may be related to improving the endogenous protective protein(HSP70)expression in exercise rats.
