摘要
采用乙醇提取法、水提取法、超声波提取法等3种不同提取方式,比较不同温度、浓度和时间下的提取效果,选定出羊栖菜多糖的最优提取工艺参数。当水浴温度90℃、提取时间3 h、浓度为1∶15时,羊栖菜多糖提取取得最佳效果。通过大孔树脂对其提取液中多糖进行静态和动态吸附、解吸处理,确定出羊栖菜多糖最佳分离纯化工艺参数:NKA-9大孔树脂进行分离工艺为吸附流速为2 BV、多糖浓度1.671 mg/mL的吸附效果最好;洗脱流速为2 BV、乙醇浓度为60%,获得羊栖菜多糖的纯度达到63.1%。
By comparing the water extraction, ethanol extraction, ultrasonic extraction method and other three kinds of extraction process under different temperature, solid-liquid ratio and time value of extracting effect, the best of the sheep food polysaccharide extraction process parameters ,when in 90 ℃ of bath temperature ,in 3 hours and solid-liquid ratio is 1∶15, the polysaccharide extraction effect is best. Then, the optimum separation and purification process parameters of polysaccharide from sheep were determined by the static and dynamic adsorption and desorption of polysaccharides in the extracted liquid by macroporous resin. In the condition of adsorption and desorption with NKA-9 macroporous resin, the adsorption effect was the best when the sample concentration was 1.671 mg/mL and the flow velocity were 2 BV. The optimum flow rate was 2 BV and the ethanol concentration was 60%. The purity of proanthocyanidin can be up to 63.1%
