摘要
以甘薯新品种徐紫薯8号为材料,研究了不同因素在诱导胚性愈伤组织和体细胞胚中的作用.结果表明:2,4-D,6-BA,KCl和蔗糖在诱导徐紫薯8号胚性愈伤中的作用大小排序为2,4-D>蔗糖>6-BA>KCl,但4个因素之间差异不显著,诱导胚性愈伤的适宜培养基为MS+2,4-D 2mg/L+蔗糖3%;NAA,6-BA,ABA和活性炭在诱导体细胞胚形成中的作用排序为NAA>6-BA>ABA>活性炭,其中NAA和6-BA为主要因素,与活性炭相比,差异显著,ABA和活性炭为次要因素,诱导体细胞胚的适宜培养基为MS+NAA 0.1mg/L+6-BA 0.2mg/L+ABA0.5mg/L+活性炭0.2%+蔗糖3%.
Using sweetpotato cultivar Xuzishu No.8 as material,the factors of influencing embryogenic callus induction and somatic embryogenesis were studied.The effect order of 2,4-D,6-BA,KCl,and sucrose on embryogenic callus induction is 2,4-D>sucrose>6-BA>KCl,but the effects among the factors are not significantly different.The effect order of NAA,6-BA,ABA,and activated carbon on somatic embryogenesis is NAA>6-BA>ABA>activated carbon,while the main factors are NAA and 6-BA,and the effects among the factors are significantly different.The best media in this study are MS+2,4-D 2 mg/L+sucrose 3%for embryogenic callus induction,and MS+NAA 0.1 mg/L+6-BA 0.2 mg/L+ABA 0.5 mg/L+activated carbon 0.2%+sucrose 3%for somatic embryogenesis.
