Effects of PP2A on Proliferation and Differentiation of Breast Cancer Stem Cell

目的探讨蛋白磷酸酶2A(protein phosphatase 2A,PP2A)在乳腺癌的表达及对乳腺癌干细胞的作用。方法运用实时定量RT-PCR检测159例乳腺浸润性癌组织和癌旁组织PP2AP的表达,构建PP2AP表达降低的乳腺癌细胞株,行球囊培养和Western-blot观察其对干细胞增殖、分化的影响。结果 PP2AP在乳腺癌组织中的表达要低于癌旁组织(P<0.05),球囊实验和western blotting提示PP2AP降低促进乳腺癌干细胞球囊生成,同时干细胞标记物表达增加。结论干扰PP2AP的表达可抑制乳腺癌干细胞的分化并促进其增殖潜能。

Objective To investigate the expressions of PP2A in breast cancer and its effect on breast cancer stem cells(BCSCs).Methods Total RNA from 159 breast cancer tissues were isolated for PP2A quantitative analysis by real-time RT-PCR.Sphere culture and Western blotting were applied to detect the proliferation and differentiation of BCSCs. Results The expression of PP2A was significantly lower in tumor tissue than that of paratumor tissue (P<0.001).Decreased expression of PP2A could increase sphere formation and expressions of stem cell makers.Conclusion Interference of PP2A expression could inhibit the differentiation of BCSCs and increase their proliferation abilities.
