白介素-35对风湿免疫性疾病诊断和治疗的影响

Effect of interleukin-35 on the diagnosis and treatment of rheumatic diseases

白细胞介素-35 (IL-35)是近年研究发现的IL-12家族的新成员,是由α链p35和β链EB病毒诱导基因3 (EBI3)组成的异源二聚体,与受体IL12Rβ2和糖蛋白130 (gp130)结合后能够激活JAK-STAT信号通路,通过调控T细胞、B细胞、树突状细胞以及巨噬细胞的分化和功能,参与多种炎性反应和自身免疫性疾病的发生发展。近年研究发现在风湿免疫性疾病,包括类风湿关节炎、系统性红斑狼疮、多发性肌炎、皮肌炎、白塞综合征、干燥综合征等,IL-35表达异常且与疾病活动度显著相关,但IL-35在风湿免疫性疾病中的确切作用仍存在争议,需进一步研究其在诊断和治疗疾病中的价值。本文就IL-35生物学特点、功能以及在风湿免疫性疾病中的研究作一综述。

Interleukin-35( IL-35),a new member of IL-12 family,is a heterodimeric cytokine composed of p35 and epstein-barr virus-inducedgene 3( EBI3) subunits. IL-35 can activate the JAK-STAT signaling pathway by binding the receptor IL12 Rβ2 and glycoprotein 130( gp130),and it is involved in the development of various inflammatory responses and autoimmune diseasesby regulating the differentiation and function of T cells,B cells,dendritic cells and macrophages. In recent years,abnormal IL-35 expression was found in rheumatic diseases,including rheumatoid arthritis,systemic lupus erythematosus,polymyositis,dermatomyositis,Beh " et’ s disease,Sjogren’s syndrome,etc. IL-35 is significantly associated with disease activity,but the exact roles of IL-35 in human rheumatic diseases remain controversial. Further studies are required to explore the value of IL-35 in diseases diagonsis and treatment. We reviewed the biological characteristics and functions of IL-35,and the research progress of IL-35 in rheumatic diseases.
