摘要
目的:研究甘肃地区乳腺导管癌妇女血浆中miR-21-5p的表达。方法:分别通过芯片法和茎环Q-PCR(Stem-loop Q-PCR)检测甘肃43名汉族乳腺导管癌患者(BC)和44名健康者(NC)血浆中的miR-21-5p水平。同时通过Stem-100P Q-PCR法测定正常乳腺上皮细胞系(HsBST-578)与四种乳腺癌细胞系中miR-21-5p的表达。结果:Q-PCR结果显示,与健康供者相比,乳腺导管癌患者血浆miR-21-5p水平显著上调(P=0.0043)。工作特征曲线(ROC)也显示了miR-21-5p对患者的显著诊断能力(AUC=0.6583,95%CI:0.5345~0.7822,P=0.0154)。miR-21-5p的血浆水平与TNM分期密切相关。乳腺癌细胞系MCF-7(5.428±1.391)中miR-21-5p的表达水平高于正常乳腺上皮细胞HsBST-578。结论:miR-21-5p在甘肃地区乳腺导管癌患者血浆中的高表达,表明miR-21-5p可以作为乳腺导管癌血浆生物标志物的潜在靶标。
Objective: We identified miR-21-5p in the plasma as a biomarker for detecting breast ductal cancer in Gansu. Methods: The plasma levels of miR-21-5p were detected in 43 patients with breast duct carcinoma and 44 healthy donors of Han women in Gansu Province by microarray and Stem-loop Q-PCR,respectively. The levels of miR-21-5P expression of HsBST-578 and four breast cancer cell lines were detemined by Stem-loop Q-PCR. Results: Q-PCR results showed the plasma miR-21-5p level were significantly up-regulated in breast ductal cancer patients compared with healthy donors(P=0.0043). Receiver operating characteristic(ROC)curve also revealed the significant diagnostic ability of mir-21-5p in patients(AUC=0.6583,95% CI: 0.5345~0.7822,P=0.0154). In addition,the plasma level of mir-21-5p was closely related to TNM staging. We also confirmed the higher expression level of miR-21-5p in breast cancer cell line MCF-7(5.428±1.391) than normal breast epithelial cell line HsBST-578. Conclusion: Taken together,miR-21-5p could represent as a potential biomarker for breast ductal cancer diagnosis in Gansu province.
