摘要
目的检测叉头框因子A2(forkhead box A2, FOXA2)和钙黏蛋白(E-cadherin)在正常结肠黏膜、结肠腺瘤及结肠腺癌组织中的表达水平,探讨其在结肠癌发生、发展过程中的作用。方法收集内蒙古医科大学第三附属医院结肠组织活检标本90例,根据病理分为正常对照组、结肠腺瘤组、结肠癌组各30例。采用实时荧光定量PCR法检测FOXA2和E-cadherin在各组织中的表达水平。结果 FOXA2、E-cadherin在正常对照组、结肠腺瘤组及结肠癌组中的相对表达水平逐渐降低,差异有统计学意义(P<0.001);在正常对照组、结肠腺瘤组及结肠癌组中FOXA2与E-cadherin的表达呈正相关(P<0.01)。结论 FOXA2和E-cadherin基因可能与结肠癌发生密切相关,通过协同作用共同作用于结肠黏膜细胞,联合检测二者的表达有助于早期诊断结肠癌,可能为基因治疗提供新靶点。
Objective To detect the expressions of forkhead box A2 (FOXA2) and E-cadherin in normal colon mucosa , colon adenoma and colon cancer tissue and investigate the values of FOXA2 and E-cadherin during the development and progression of colon cancer. Methods All specimens were collected from the Department of Gastroenterology outpatients or inpatients at the Third Affiliated Hospital of Inner Mongolia Medical University. Colonoscopy was performed to obtain colonic mucosa specimens during colonoscopy or treatment and combined with pathological diagnosis of normal control group, colon cancer group, colon adenoma group, each group consist of 30 cases. Real-time fluorescence quantitative PCR was used to detect the expression of FOXA2 and E-cadherin in normal colon mucosa, colon adenoma and colon cancer tissues. Results The trend of expressions of FOXA2 and E-cadherin in normal colon mucosa, colon adenoma and colon cancer tissues gradually decreased, the difference was statistically significant ( P <0.001);in normal control group, colon adenoma group and colon cancer group, the expression of FOXA2 was positively correlated with E-cadherin ( P <0.01). Conclusion FOXA2 and E-cadherin genes may be closely related to the development of colon cancer, and act synergistically on colonic mucosa cells. The combined detection of both may help early diagnosis of colon cancer and may provide a new target for gene therapy.
