摘要 目的观察扭曲因子Twist在宫颈癌细胞株Hela中的表达变化,并探讨其对Hela细胞侵袭转移的影响以及作用机制。方法取宫颈癌细胞株Hela以及人宫颈永生化上皮细胞H8,采用Western blotting法检测细胞中Twist蛋白。常规培养宫颈癌细胞株Hela,分为对照组、过表达组、沉默组,后两组采用慢病毒转染技术分别过表达和沉默细胞中的Twist基因,采用boyden实验检测细胞侵袭能力;细胞划痕实验检测细胞迁移能力;Western blotting法检测Rac1以及MMP-2蛋白。结果 Hela细胞中Twist蛋白表达高于H8细胞(P<0.05)。与对照组相比,过表达组细胞中Twist蛋白表达升高,细胞侵袭、迁移能力增强,Rac1、MMP-2蛋白表达增加(P均<0.05);与对照组相比,沉默组细胞中Twist蛋白表达降低,细胞侵袭、迁移能力减弱,Rac1、MMP-2蛋白表达降低(P均<0.05)。结论 Twist在宫颈癌细胞株Hela中高表达,促进/抑制其表达可以显著增强/降低Hela的侵袭转移,机制可能是通过作用于侵袭转移相关蛋白Rac1、MMP-2的表达进而调控细胞的侵袭转移。 Objective To observe the expression of Twist in the cervical cancer Hela cells, and to explore its effect on the invasion and metastasis of Hela cells and its mechanism. Methods Cervical cancer cell line Hela and immortalized human cervical epithelial cell H8 was obtained, the Twist protein in cells was detected by Western blotting. The cervical cancer cells Hela underwent routine cultured, and were divided into the control group, overexpression group, and silent group. The Twist gene was overexpressed and silenced by lentivirus transfection in the latter two groups, and the cell invasive ability was detected by boyden assay;the cell migration ability was detected by cell Scratch assay;the expression of Rac1 and MMP-2 protein was detected by Western blotting. Results The expression of Twist protein in Hela cells was higher than that in H8 cells ( P <0.05). Compared with the control group, the expression of Twist protein increased, the invasion and migration abilities of cells increased, and the expression of Rac1 and MMP-2 protein increased in the overexpression group (all P <0.05). Compared with the control group, the expression of Twist protein and rac1 and MMP-2 protein decreased in the silent group (all P <0.05). Conclusion Twist is highly expressed in cervical cancer Hela cells, promoting/inhibiting Twist's expression can significantly enhance/decrease Hela's invasion and metastasis by acting on the expression of invasion and metastasis-related proteins Rac1 and MMP-2.
