中药方剂胃肠舒组方及其成分药灌胃SD大鼠的胃肠动力观察 被引量:3

Observation of gastric and intestinal dynamics of SD rats after administration of Chinese herbal medicine weichangshu pills and its ingredients

摘要 目的观察中药方剂胃肠舒组方及其成分药物灌胃对SD大鼠胃肠动力的影响。方法 80只SD大鼠随机分为空白组、胃肠舒组、胃肠舒成分药组(大黄白芍组、大黄甘草组、白芍甘草组、白芍组、大黄组和甘草组),每组各10只。适应性喂养3天后取各组大鼠,分别用蒸馏水,75 mg/m L的大黄、白芍及甘草灌胃液,50 mg/m L的大黄、白芍灌胃液,50 mg/m L的大黄、甘草灌胃液,50 mg/m L的白芍、甘草灌胃液,25 mg/m L的白芍灌胃液,25 mg/m L的大黄灌胃液,25 mg/m L的甘草灌胃液灌胃。20 m L/kg一天一次,连续灌胃7 d。灌胃第7天时,各组SD大鼠禁食(不禁水、药) 18 h,末次灌胃40 min时予20 mg/m L的葡聚糖蓝(BD) 2000溶液0. 4 m L灌胃后颈椎脱臼处死,剖腹取全胃,测算各组大鼠胃动力(胃内色素排空率表示)、肠动力(肠推进率)。结果胃肠舒组、白芍组、大黄组、甘草组、大黄白芍组、大黄甘草组、白芍甘草组及空白组胃内色素排空率分别为94. 44%±1. 95%、88. 73%±6. 80%、89. 97%±3. 09%、91. 92%±2. 51%、87. 38%±5. 88%、82. 98%±12. 58%、9. 80%±7. 55%、88. 29%±6. 63%。与其余各组比较,甘草组、胃肠舒组大鼠胃内色素排空率升高(P <0. 05)。甘草对胃排空有单独效应(P<0. 05),白芍、大黄与甘草配伍对胃排空存在交互作用(P <0. 05)。在促进胃排空方面,白芍、大黄、甘草三味药有协同交互作用。胃肠舒组、白芍组、大黄组、甘草组、大黄白芍组、大黄甘草组、白芍甘草组及空白组大鼠肠推进率分别为41. 35%±6. 69%、53. 39%±8. 32%、53. 34%±6. 23%、56. 10%±6. 62%、61. 21%±7. 80%、50. 635±10. 59%、63. 06%±7. 96%、62. 35%±4. 78%,与其他各组比较,白芍、大黄、甘草、大黄白芍组大鼠肠推进率升高(P均<0. 05)。大黄、白芍、甘草对肠推进有单独效应(P均<0. 05),白芍、大黄配伍对肠推进有交互作用(P <0. 05)。白芍、大黄配伍对肠推进有协同交互作用。结论胃肠舒组方灌胃可提高SD大鼠的胃、肠动力。 ObjectiveTo observe the effects of Chinese herbal medicine Weichangshu pills and its ingredients on gastric and intestinal dynamics in SD rats.MethodsEighty SD rats were randomly divided into the blank group,Weichangshu pills group,and Weichangshu pills ingredients group[rhubarb and paeony(RP)group,rhubarb and licorice(RL)group,paeony and licorice(PL)group,paeony(P)group,rhubarb(R)group,and licorice(L)group],with 10 rats in each group.After 3 days of adaptive feeding,rats in each group were respectively given distilled water,75 mg/mL of rhubarb,paeony and liquorice for gastric irrigation,50 mg/mL of rhubarb and paeony for gastric irrigation,50 mg/mL of rhubarb and liquorice for gastric irrigation,50 mg/mL of paeony and liquorice for gastric irrigation,25 mg/mL of paeony for gastric irrigation,25 mg/mL of rhubarb for gastric irrigation,and 25 mg/mL of liquorice for gastric irrigation,20 mL/kg once a day,and for 7 d.On the 7th day of gavage,SD rats in each group fasted(water and medicine could not be prevented)for 18 h,and 20 mg/mL of dextran blue(BD)2000 solution was given to 0.4 mL at the last gavage for 40 min,and then the cervical vertebra was dislocated and sacrificed.The whole stomach was taken by laparotomy,and the gastric motility(represented by the rate of gastric pigment emptying)and intestinal motility(the rate of intestinal propulsion)were measured.ResultsThe gastric emptying rates in the Weichangshu pills group,paeony group,rhubarb group,licorice group,rhubarb and paeony group,rhubarb and licorice group,paeony and licorice group and blank group were 94.44%±1.95%,88.73%±6.80%,89.97%±3.09%,91.92%±2.51%,87.38%±5.88%,82.98%±12.58%,9.80%±7.55%,and 88.29%±6.63%.Compared with the other groups,the gastric emptying rates of rats in the licorice group and the Weichangshu pills group increased(both P<0.05).Licorice had a separate effect on gastric emptying(P<0.05).The compatibility of paeony,rhubarb and licorice had an interaction effect on gastric emptying(P<0.05).Paeony,rhubarb and liquorice had synergistic interaction in promoting gastric emptying.The intestinal propulsion rates of rats in the Weichangshu pills group,paeony group,rhubarb group,licorice group,rhubarb and paeony group,rhubarb and licorice group,paeony and licorice group and blank group were 41.35%±6.69%,53.39%±8.32%,53.34%±6.23%,56.10%±6.62%,61.21%±7.80%,50.635±10.59%,63.06%±7.96%,and 62.35%±4.78%;compared with the other groups,the intestinal propulsion rate of rats in the groups of paeony,rhubarb,liquorice and rhubarb and paeony increased(all P<0.05).Rhubarb,paeony and liquorice had separate effects on intestinal propulsion(all P<0.05),while paeony and rhubarb had an interactive effect on intestinal propulsion(P<0.05).Paeony and rhubarb had synergistic and interactive effects on intestinal propulsion.ConclusionsThe gastric motility and intestinal motility of SD rats can be improved by gavage of Weichangshu pills.
