摘要
建立黑豆皮中原花青素的含量测定方法,并且通过ABTS+清除试验来测试抗氧化活性。采用HPLC法测定黑豆皮中原花青素的含量,色谱柱采用Agilent Eclipse XDB-C18柱(250 mm×4.60 mm,5μm),流动相A相为2%冰醋酸水溶液,B相为甲醇,进行梯度洗脱,流速1.0 m L/min,检测波长280 nm,柱温30℃,进样量20μL。结果表明,在5~500μg范围内线性关系良好,最低检出限1.0μg/m L,平均加样回收率为101.43%。黑豆皮中的原花青素的平均含量为104.14μg/g。体外抗氧化试验表明,黑豆皮中的原花青素对应的IC50值为13.62μg/m L,远小于维生素C 98.23μg/m L。该方法简便、快速、准确,具有良好的重复性和回收率,黑豆皮中原花青素具有较高的抗氧化活性。
To establish a method for determination of proanthocyanidins(PC)content in black soybean hulls.In addition,the antioxidant activity of extracts from black soybean hulls in vitro were tested by ABTS+.To develop an RP-HPLC method for the determination of the proanthocyanidins in black soybean hulls.The chromatographic separation was performed on Agilent Eclipse XDB-C18(250 mm×4.60 mm,5μm)column by gradient elution using 2%glacial acetic acid solution as mobile phase A and methanol as mobile phase B at a flow rate of 1.0 mL/min.The column temperature was set at 30℃and the detection wavelength at 280 nm.The injection volume was 20μL.The results showed that the linearity of proanthocyanidins was 5~500μg,the detection limit was 1.0μg/mL,and the average recovery rate was 101.43%,respectively.The contents of proanthocyanidins in black bean hulls was 104.14μg/g.In the radical scavenging assay,the IC50 values of black bean hulls was 13.62μg/mL,which were lower than those of Vitamin C(98.23μg/mL).This method is simple,reproducible,accurate and reliable and the content of proanthocyanidins from black bean hulls have higher reducing power.
