摘要
目的探讨多巴胺D受体基因多态性与抗精神病药物疗效之间的相关性。方法选择2015年8月—2016年8月期间该院收治的46例精神分裂症患者为研究对象,均采用氯氮平治疗,运用PANSS量表评价氯氮平疗效,并且运用测定基因序列对多巴胺受体D上3个SNP位点的等位基因频率和基因型进行检测。结果该组50例患者氯氮平血药浓度范围为167~585 ng/mL,平均(363.2±97.1)ng/mL,并且氯氮平有效组的rs265981基因型CC、TC、TT分布的平均血药浓度分别为(358.8±101.4)、(381.9±87.5)、(352.1±0.0)ng/mL;同时,相比较氯氮平无效组而言,有效组的等位基因T、C以及rs265981基因型TT、TC以及CC分布较广,组间对比差异有统计学意义(P<0.05)。结论多巴胺D受体基因rs5326、rs265981与抗精神病药物的治疗效果有关。
Objective To investigate the relationship between dopamine D receptor gene polymorphism and antipsychotic efficacy. Methods 46 patients with schizophrenia admitted to our hospital from August 2015 to August 2016 were enrolled in the study. All patients were treated with clozapine. The PANSS scale was used to evaluate the efficacy of clozapine and the gene sequence was determined. The allele frequencies and genotypes of the three SNP sites on dopamine receptor D were detected. Results The concentration of clozapine in 50 patients in this group ranged from167-585 ng/mL, with an average of(363.2±97.1) ng/mL, and the rs265981 genotype CC, TC, TT in the clozapine-effective group. The mean plasma concentrations of the distribution were(358.8±101.4),(381.9±87.5), and(352.1±0.0) ng/mL,respectively. Meanwhile, the allele T, C of the effective group was compared with the clozapine-ineffective group. The TT, TC and CC of the C, rs265981 genotypes were widely distributed, and the differences between the groups were significant(P<0.05). Conclusion The dopamine D receptor genes rs5326 and rs265981 are related to the therapeutic effects of antipsychotics.
